摘 要:目的 探讨异叶青兰总黄酮对自发性高血压大鼠肾损伤及磷脂酰肌醇-3-羟激酶(PI3K)/蛋白质激酶B(AKT)/雷帕霉素靶蛋白(mTOR)信号通路影响。方法 将10只WKY大鼠作为对照组,40只雄性自发性高血压大鼠(SHR大鼠)按照随机数字表分为模型组(等体积蒸馏水)、低剂量实验组(300 mg·kg^(-1)异叶青兰总黄酮)、高剂量实验组(600 mg·kg^(-1)异叶青兰总黄酮)、阳性对照组(0.67 mg·kg^(-1)依那普利)。各组大鼠每天灌胃1次,连续灌胃8周。末次给药后,测量血压、收集24 h尿液。用过碘酸-雪夫(PAS)法检测各组大鼠肾组织病理学变化;用酶联免疫吸附实验(ELISA)试剂盒检测4 h尿液微量白蛋白(mAlb)含量;用蛋白质印迹法检测各组大鼠肾组织中PI3K/AKT/mTOR信号通路相关蛋白表达情况。结果 对照组、模型组、高剂量实验组、阳性对照组大鼠给药8周后收缩压分别为(113.71±7.01)、(179.85±13.22)、(145.94±7.26)和(141.55±7.35)mmHg;mAlb含量分别为(18.78±2.52)、(34.34±2.67)、(24.23±2.02)和(23.96±1.62)μg·mL^(-1);p-PI3K蛋白水平分别为1.04±0.08、0.32±0.04、0.78±0.08和0.75±0.09。以上指标,对照组与模型组比较,高剂量实验组、阳性对照组与模型组比较,差异均有统计学意义(均P<0.05)。结论 异叶青兰总黄酮可减缓自发性高血压大鼠肾损伤,这可能与激活PI3K/AKT/mTOR信号通路有关。Objective To investigate the effects of dracocephalum heterophyllum benth flavone on renal injury and phosphatidylinositol 3-hydroxykinase(PI3K)/protein kinase B(AKT)/target of rapamycin(mTOR)signaling pathway in spontaneously hypertensive rats.Methods Ten WKY rats were selected as control group,and 40 male spontaneous hypertension rats(SHR)rats were randomly divided into model group(equal volume of distilled water),experimental-L group(300 mg·kg^(-1)dracocephalum heterophyllum benth flavone),experimental-H group(600 mg·kg^(-1)dracocephalum heterophyllum benth flavone),positive control group(0.67 mg·kg^(-1)enalapril).Blood pressure was measured and urine was collected for 24 h after the last administration.The pathological changes of renal tissue in each group were detected by periodic acid-schiff(PAS)method.The levels of urine microalbumin(mAlb)and N-acetyl-β-D-glucosidase(NAG)were detected by enzyme-linked immunosorbent assay(ELISA)kit.The expression of PI3K/AKT/mTOR signaling pathway related proteins in renal tissue of rats in each group was detected by Western bloting.Results The systolic blood pressure of rats in control,model,experimental-H and positive control groups after 8 weeks of administration was(113.71±7.01),(179.85±13.22),(145.94±7.26)and(141.55±7.35)mmHg,respectively;mAlb contents were(18.78±2.52),(34.34±2.67),(24.23±2.02)and(23.96±1.62)μg·mL^(-1),respectively;p-PI3K protein levels were 1.04±0.08,0.32±0.04,0.78±0.08 and 0.75±0.09,respectively.The above indexes showed statistically significant differences between control group and model group,between experimental-H group,positive control group and model group(all P<0.05).Conclusion Dracocephalum heterophyllum benth flavone can alleviate renal injury in spontaneously hypertensive rats,which may be related to the activation of PI3K/AKT/mTOR signaling pathway.
