摘  要:目的建立半枝莲高效液相色谱(high performance liquid chromatography,HPLC)指纹图谱联用化学计量学的质量分析方法。方法用InertSustain C_(18)(250 mm×4.6 mm,5μm),乙腈-1 mL·L^(-1)磷酸溶液梯度洗脱,梯度流速,进样量为10μL,波长为335 nm,柱温为30℃,用相似度评价法、聚类分析、主成分分析(principle component analysis,PCA)和偏最小二乘法判别分析(partial least squares discriminant analysis,PLS-DA)评价。结果12批样品相似度均大于0.90;聚类分析河南为一类,云南为一类,广东、广西为一类;PCA分类结果与聚类分析结果一致,差异大的色谱峰有10个;PLS-DA分类结果与聚类分析结果一致,差异大的色谱峰有14个;筛选半枝莲潜在质量标志物的色谱峰有7个,为10、11(木犀草苷)、12(野黄芩苷)、14、16、18、19号峰。结论建立了半枝莲高效液相色谱指纹图谱联用化学计量学的质量分析方法,为半枝莲质量标准研究提供依据。Objective To establish a quality control method of Scutellaria barbata D.Don by hiph performance liquid chromatography(HPLC)fingerprints combined with chemometrics.Methods An InertSustain C_(18) column(250 mm×4.6 mm,5μm)was used with acetonitrile 1 mL·L^(-1) phosphoric acid solution as the mobile phase for gradient elution.The injection volume was 10μL,the detection wavelength was 335 nm,and the column temperature was 30℃.The similarity,cluster analysis,principal component analysis(PCA)and partial least squares discriminant analysis(PLS-DA)were used for evaluation.Results The similarity of 12 batches of samples was higher than 0.90.The samples were divided into He nan,Yunnan,Guangdong and Guangxi habitats by cluster analysis,similar to the results of PCA classification with 10 different chromatographic peaks,and also similar to the results of PLS-DA classification with 14 different chromatographic peaks.Seven chromatographic peaks,i.e.10,11(luteolin),12(baicalin),14,16,18 and 19 were screened as the quality markers of Scutellaria barbata D.Don.Conclusion The quality control method of Scutellaria barbata D.Don fingerprints combined with chemometrics was established,which provides a basis for the study of quality standard of Scutellaria barbata D.Don.
