摘 要:目的明确前列消调节低氧诱导因子1-α(HIF1-α)、血管内皮生长因子(VEGF)治疗前列腺增生(BPH)模型大鼠的作用及机制。方法将60只Wistar大鼠随机分为正常组、模型组、前列消高、中、低剂量组和西药组。除正常组外,其他各组以手术去势后皮下注射丙酸睾酮(5 mg·kg^(-1)·d^(-1)),连续30 d,建立前列腺增生模型。造模完成后,相应给药治疗21 d。取材大鼠前列腺组织,HE染色法观察前列腺组织增生病理变化;免疫组化、免疫荧光法检测血小板-内皮细胞黏附分子(CD31)、血管内皮生长因子受体1(VEGFR1)表达评估血管新生;Western blot检测低氧诱导因子1-α(HIF1-α)、VEGF caspase表达量,RT-PCR检测大鼠前列腺组织HIF1-α、VEGF caspase-9 mRNA的表达量。结果与正常组比较,模型组前列腺体积显著性增大,CD31、VEGFR1含量增加,提示腺体内有明显血管增生;经治疗后,前列消高、中剂量组及西药组前列腺体积缩小,降低CD31、VEGFR1含量,HIF1-α、VEGFR caspase-9蛋白及mRNA表达显著性降低。结论前列消改善丙酸睾酮诱导的大鼠前列腺增生,其机制可能与HIF1-α/VEGFR通路有关。Objective To investigate the effect and mechanism of Qianlie Xiao decoction treats benign prostatic hyperplasia(BPH)in rats by regulating hypoxia-inducible factor 1-α(HIF1-α)and vascular endothelial growth factor(VEGF).Meth⁃ods Sixty Wistar rats were randomly divided into normal group,model group,Qianliexiao decoction high-,medium-,low-dose group and western medicine group.Except for the normal group,the BPH model was established by subcutaneous injection of testosterone propionate(5 mg·kg^(-1)·d^(-1))after surgical castration for 30 days.After the model was established,the patients were given corresponding drugs for 21 days.HE staining was used to observe the pathological changes of prostate hyperplasia.The expression of platelet-endothelial cell adhesion molecule(CD31)and vascular endothelial growth factor receptor1(VEGR1)was detected by immunohistochemistry and immunofluorescence to evaluate angiogenesis.The expressions of hypoxia-inducible factor 1-α(HIF1-α),VEGFR1 caspase-9 were detected by Western blot,and the mRNA expressions of HIF1-α,VEGF and caspase-9 in prostate tissue were detected by RT-PCR.Results Compared with the normal group,the prostate volume of the model group was significantly increased,and the content of CD31 and VEGFR1 was increased,suggesting that there was obvious vascular hyperplasia in the gland.After treatment,the prostate volume was reduced,CD31 and VEGFR1 contents were de⁃creased,and HIF1-α,VEGF caspase-9 protein and mRNA expressions were significantly decreased in the high and medium dose groups and western medicine groups.Conclusion Qianliexiao decoction can improve prostatic hyperplasia induced by testos⁃terone propionate in rats,and its mechanism may be related to HIF1-α/VEGF pathway.
