摘  要:目的探讨芍药甘草汤对腹泻型肠易激综合征(IBS-D)小鼠结肠组织BDNF及其受体TrkB的表达及对内脏高敏性的影响。方法将60只ICR小鼠随机分为空白组、模型组、匹维溴铵组、芍药甘草汤低、中、高剂量组,除空白组外,其余各组均采用急慢性刺激+番泻叶液体灌胃诱导IBS-D模型;分别观察大便含水率以及腹壁撤退反射情况;结肠组织病理形态改变以及结肠组织BDNF、TrkB蛋白表达水平。结果与空白组比较,模型组小鼠一般情况恶化,体质量增长缓慢(P<0.05),大便含水率上升(P<0.05),AWR评分升高(P<0.05)。与空白组比较,模型组BDNF、TrkB蛋白表达水平显著上升(P<0.01);与模型组比较,各给药组IBS小鼠的一般状况好转,体质量上升(P<0.05),大便含水率下降(P<0.05)、AWR评分下降(P<0.05),各治疗组小鼠结肠组织蛋白BDNF表达均有下降(P<0.05),其中芍药甘草汤高剂量组BDNF蛋白表达水平与空白组相近(P<0.01)。芍药甘草汤中、高剂量组与西药阳性对照组TrkB蛋白表达下降,其中与芍药甘草汤高剂量组最为明显,并呈剂量依赖(P<0.01)。结论芍药甘草汤可通过减轻结肠组织中的BDNF、TrkB信号通路的表达,减轻小鼠内脏高敏感性,从而缓解腹泻型肠易激综合征小鼠腹痛腹泻的程度,发挥治疗腹泻型肠易激综合征的作用。Objective To investigate the effect of Shaoyao Gancao Decoction on the expression of BDNF and its receptor TrkB in colon tissue of mice with diarrhea-predominant irritable bowel syndrome(IBS-D)and on visceral hypersensitivity.Methods 60 ICR mice were randomly divided into normal group,model group,pinaverium bromide group,Shaoyao Gancao de⁃coction low-dose,medium-dose and high-dose groups.Except for the normal group,other groups were treated with acute and chronic stimulation of senna.The IBS-D model was induced by gavage;the stool water content and abdominal wall withdrawal reflex;the pathological changes of colon tissue and the expression levels of BDNF and TrkB in colon tissue were observed.Results Compared with the normal group,the general condition of the mice in the model group deteriorated,the body weight increased slowly(P<0.05),the water content of the stool increased(P<0.05),and the AWR score increased(P<0.05).Compared with the blank group,the protein expression levels of BDNF and TrkB in the model group increased significantly(P<0.01);compared with the model group,the general condition of the IBS mice in each administration group improved,the body weight in⁃creased(P<0.05),and the water content of the stools.The expression of BDNF in the colon tissue of mice in each treatment group decreased(P<0.05),the AWR score decreased(P<0.05),and the expression level of BDNF protein in the high-dose Shaoyao Gancao Decoction group was similar to that in the blank group(P<0.01).The expression of TrkB protein decreased in the middle and high dose groups of Shaoyao Gancao Decoction and the western medicine positive control group,which was the most obvious in the high dose group of Shaoyao Gancao Decoction,and showed a dose-dependent manner(P<0.01).Conclusion The expression of BDNF and TrkB signaling pathways can reduce the hypersensitivity of viscera in mice,thereby reducing the degree of abdominal pain and diarrhea in mice with diarrhea-type irritable bowel syndrome,and play a role in the treatment of diarrhea-typ
