左归丸母代干预对先天肾虚仔鼠脑皮质层发育及ADAMTS-4、Reelin、Fyn调控因子的影响

摘　　要：目的 研究左归丸干预Reelin信号通路上下游相关因子在脑皮质层发育过程的作用及机制。方法 观察第21日龄仔鼠的一般情况、测量体重、体长和脑重量;观察脑皮质层发育状况;检测ADAMTS-4、Reelin和Fyn蛋白和mRNA的表达。结果 空白对照组仔鼠各方面情况较好,模型组皮肤颜色偏暗,易惊恐、小便次数增多、大便稀软,补肾组皮肤颜色、二便正常。与空白对照组比较,模型组体重、体长及脑重量减少趋势;与模型组相比,补肾组呈有所好转。与对照组比较,模型组ADAMTS-4蛋白和mRNA表达量明显升高,Reelin、Fyn蛋白和mRNA表达量明显降低;与模型组比较,补肾组ADAMTS-4蛋白和mRNA表达量降低,Reelin、Fyn蛋白和mRNA表达量升高。结论 先天肾虚仔鼠幼儿期学习记忆能力下降、生长发育迟缓,左归丸填补母鼠肾精,能抑制脑皮质层ADAMTS-4表达,增强Reelin表达和活性,激活Fyn下游通路,促进脑皮质发育。Objective To study the effect and mechanism of Zuoguiwan on the development of cerebral cortex by interfering the upstream and downstream related factors of Reelin signaling pathway.Methods The general condition,body weight,body length and brain weight of 21-day-old pups were observed,the development of cerebral cortex was observed,and the protein and mR⁃NA expressions of ADAMTS-4,Reelin and Fyn were detected.Results The blank control group was better than the model group in all aspects.The model group's skin color was dark,easy to panic,frequent urination,loose stool,and the kidney-tonifying group's skin color and stool were normal.Compared with the blank control group,the body weight,body length and brain weight of the model group decreased,and compared with the model group,the kidney-tonifying group showed an increasing trend.Compared with the control group,the expression levels of ADAMTS-4 protein and mRNA in the model group were significantly increased,while the expression levels of Reelin,Fyn protein and mRNA were significantly decreased,and the expression levels of ADAMTS-4 protein and mRNA in the kidney-tonifying group were significantly decreased as compared with the model group,the expression of Reelin,Fyn protein and mRNA were increased.Conclusion The Learning and memory ability of the pups with congenital kidney deficiency is decreased and their growth and development is retarded.Zuogui pill can inhibit the expression of ADAMTS-4,enhance the expression and activity of Reelin,and activate the downstream pathway of Fyn,promote the develop⁃ment of cerebral cortex.
