摘 要:目的:探讨电导率-含水量曲线法的可靠性,并使用该方法确定O/W型微乳成型临界点,制备及优化广藿香挥发油微乳。方法:比较目测法和电导率-含水量曲线法确定O/W型微乳成型临界点的精确性;伪三元相图筛选油相、表面活性剂、助表面活性剂的类型、Km值;以微乳的平均粒径、多分散系数(PDI)作为评价指标,响应面法优化广藿香挥发油微乳的配制条件;优化后的微乳分别遮光、不遮光、高速离心等方法考察其稳定性。结果:电导率-含水量曲线法确定的O/W型微乳成型临界点更为精确;伪三元相图筛选出的微乳处方为m广藿香挥发油/mIPM=(1:1)、TW80、PEG400;响应面法筛选出的最优处方为:广藿香挥发油为10.07%,IPM为10.07%,TW80为60.43%,PEG400为19.43%,Km值为3.11,搅拌速率为622 r/min,该工艺下制得的微乳的平均粒径为(17.68±0.47)nm、PDI为(0.074±0.003);优化后的微乳稳定性较好。结论:电导率-含水量曲线法确定的O/W型微乳成型临界点精确性高,使用该方法筛选、优化微乳后,获得的广藿香挥发油微乳稳定性好。Objective:To explore the reliability of the conductivity-water content curve method,and use this method to determine the critical point of O/W microemulsion formation,preparation and optimization of pogostemon cablin volatile oil microemulsion.Methods:The accuracy of visual method and conductivity-water content curve method for determining the critical point of O/W microemulsion forming was compared.The pseudo-ternary phase diagram was used to screen the type and Km value of oil phase,surfactant and cosurfactant.The average particle size and polydispersity index(PDI)of microemulsion were used as evaluation indexes,and the preparation conditions of pogostemon cablin volatile oil microemulsion were optimized by response surface method.The stability of the optimized microemulsion was investigated by shading,non-shading and high-speed centrifugation.Results:The critical point of O/W microemulsion forming determined by conductivity-water content curve method was more accurate.The microemulsion prescription screened by pseudo-ternary phase diagram was mpogostemon cablin oil/mIPM=(1:1),TW80,PEG400.The optimal prescription screened by response surface methodology was as follows:pogostemon cablin volatile oil was 10.07%,IPM was 10.07%,TW80 was 60.43%,PEG400 was 19.43%,Km was 3.11,and the stirring rate was 622 r/min.The average particle size of the microemulsion prepared by this process was(17.68±0.47)nm,and the PDI was(0.074±0.003).The optimized microemulsion has good stability.Conclusion:The accuracy of the critical point of O/W microemulsion determined by the conductivity-water content curve method is high.After screening and optimizing the microemulsion by this method,the obtained pogostemon cablin volatile oil microemulsion has good stability.
