摘  要:目的探讨H_(2)S诱导胃黏膜细胞(gastric mucosal epithelial cells,GES-1)凋亡的作用机制。方法在不同浓度的H_(2)S合成酶胱硫醚-γ-裂解酶(cystathionine-γ-lyase,CSE)抑制剂炔丙基甘氨酸(propargylglycine,PAG)的作用下,外源性H_(2)S供体NaHS(400μmol/L)作用于胃黏膜细胞GES-1。通过AnnexinV-PE/7-AAD双标染色法利用MUSE细胞状态分析仪检测细胞凋亡率,蛋白质印迹检测Caspase-3表达。结果与对照组比较,NaHS组GES-1细胞凋亡率增高(P<0.001)、Caspase-3激活(P<0.001);与NaHS组比较,NaHS-PAG 100μmol/L组的早期凋亡率降低(P<0.001),Caspase-3断裂带(相对分子质量约为17×10^(3))表达降低(P<0.001);而NaHS-PAG 500μmol/L组及NaHS-PAG 1000μmol/L组细胞晚期凋亡率都增高(P均<0.001),Caspase-3断裂带表达都增加(P=0.003、P<0.001),差异有统计学意义。结论PAG双向调控NaHS诱导胃黏膜细胞GES-1凋亡。Objective To explore the effect of H_(2)S on apoptosis of the gastric mucosal cells(GES-1).Methods In the presence of different concentrations of the cystathionine-γ-lyase(CSE)inhibitor propargylglycine(PAG),GES-1(400μmol/L)cells were treated with exogenous H_(2)S donor NaHS.The apoptotic rate was detected by the AnnexinV-PE/7-AAD double-labeling method,the expression of Caspase-3 was determined by Western-blotting.Results Compared with the control group,the apoptosis rate of GES-1 cells and the activation of caspase-3 in NaHS group were significantly higher(all P<0.001).Compared with the NaHS group,the early apoptosis rate of the NaHS-PAG 100μmol/L group weakened and the expression of Caspase-3(relative molecular weight is 17×10^(3))decreased with statistical significance(all P<0.001),while the late apoptosis rate of the NaHS-PAG 500μmol/L group and the NaHS-PAG 1000μmol/L group increased with statistical significance(all P<0.001),and the expression of Caspase-3(relative molecular weight is 17×10^(3))increased(P=0.003,P<0.001),the difference was statistically significant.Conclusion The results show that PAG bidirectional regulates NaHS-induced apoptosis of GES-1 cells.
