复方前列康胶囊治疗慢性前列腺炎的活性作用研究

摘　　要：目的探讨复方前列康胶囊治疗慢性前列腺炎的活性作用研究。方法采用高效液相色谱法对复方前列康胶囊提取物进行成分分析;通过分析复方前列康胶囊提取物对DPPH自由基、ABTS自由基、羟基自由基和O2-·的清除能力来测定体外抗氧化活性;通过建立角叉菜胶致大鼠足肿胀模型观察其抗炎作用;应用ELISA法检测前列腺组织中白细胞和卵磷脂小体数目、促炎因子COX-2、iNOS及炎症因子TNF-α、IL-1β、IL-6、IL-8、PEG2的含量;利用qRT-PCR方法检测前列腺组织中PI3K、Akt、RAGE mRNA表达。结果化学成分分析表明主要含有芦丁、槲皮素、黄柏碱和盐酸小檗碱等成分。抗氧化实验结果表明复方前列康胶囊提取物具有优良的体外抗氧化能力。抗炎实验表明,复方前列康胶囊提取物可以抑制角叉菜胶所致的小鼠足肿胀。前列腺炎活性实验结果表明,复方前列康胶囊提取物可以明显的降低大鼠前列腺液中白细胞数目,提高卵磷脂小体密度,降低大鼠前列腺组织中促炎因子COX-2、iNOS及炎症因子TNF-α、IL-1β、IL-6、IL-8、PEG2的含量。qRT-PCR结果显示,复方前列康胶囊提取物可以下调大鼠前列腺组织中PI3K、Akt、RAGE mRNA表达含量。结论复方前列康胶囊提取物可能通过PI3K-Akt和AGE-RAGE两条通路、多成分协同作用的方式抑制炎症因子的生成以及减轻氧化应激反应,进而对慢性前列腺炎起到治疗作用。Objective To study the active action of compound Qianliekang capsules in the treatment of chronic prostatitis.Methods The components of extract of compound Qianliekang capsules were analyzed by high performance liquid chromatography.The antioxidant activity in vitro was determined by analyzing the scavenging ability of compound Qianliekang capsule extract on 1,1-Diphenyl-2-picylhydrazyl(DPPH)free radicals,2,2'-Azinobis-(3-ethylbenzthiazoline-6-sulphonate)(ABTS)free radicals,hydroxyl free radicals,and Tris-HCl(O_(2)^(-)•).The anti-inflammation effect of carrageenan was observed by establishing carrageenan induced rat foot swelling model.The number of white blood cells and phospholipid bodies,and the contents of proinflammatory factors Cyclooxygenase-2(COX-2)and inducible nitric oxide synthase(iNOS),and inflammatory factors tumor necrosis factor(TNF-α),IL(interleukin,IL)-1β,IL-6,IL-8 and prostaglandin E2(PEG2)in prostate tissues were detected by enzyme linked immunosorbent assay(ELISA).The expressions of phosphatidylinositol 3-kinase(PI3K),Akt and receptor for advanced glycation end products(RAGE)mRNA in prostate tissues were detected by quantitative real time polymerase chain reaction(qRT-PCR).Results Chemical composition analysis showed that the extracts of compound Qianliekang capsule mainly contained rutin,quercetin,phellodendron and berberine hydrochloride.The results of antioxidant experiment showed that the extracts of compound Qianliekang capsule had excellent antioxidant ability in vitro.The anti-inflammatory experiment showed that the compound Qianliekang capsule extracts could inhibit the rat foot swelling caused by carrageenan.The results of prostatitis activity experiment showed that compound Qianliekang capsule extracts could significantly reduce the number of white blood cells in the prostatic fluid of rats,increase the density of phospholipid bodies,and reduce the contents of proinflammatory factors COX-2,iNOS,and inflammatory factors TNF-α,IL1β,IL-6,IL-8 and PEG2 in the prostate tissue of rat
