干旱胁迫下油用牡丹‘凤丹’基因组DNA甲基化分析

摘　　要：利用甲基化敏感扩增多态性技术探究不同干旱处理下油用牡丹‘凤丹’基因组DNA甲基化的水平及其变异模式。结果表明:‘凤丹’4年生植株的基因组DNA甲基化率为93.24%;随着干旱程度的加强,‘凤丹’基因组DNA发生甲基化变化位点的比率呈先上升后下降的趋势;对不同程度干旱处理后的‘凤丹’进行复水,与对照植株相比,复水后的‘凤丹’植株基因组DNA总甲基化率均表现为上升。干旱胁迫下‘凤丹’基因组DNA甲基化出现4种变异模式,表现为15种条带类型。轻度干旱诱导‘凤丹’基因组39.78%的位点发生了甲基化,40.00%的位点发生了去甲基化;重度干旱诱导‘凤丹’基因组46.00%的位点发生了甲基化,38.60%的位点发生了去甲基化,表明干旱胁迫会诱导‘凤丹’基因组DNA甲基化状态发生改变。The methylation sensitive amplification polymorphism(MSAP) technique was used to explore thegenome DNA methylation levels and variation patterns of oil tree peony ( Paeonia ostii) ‘ Fengdan’ underdifferent drought treatments. The results showed that the genome DNA methylation rate of P. ostii ‘Fengdan’4-year-old plants was 93. 24%. With the intensification of drought,the proportion of loci where genomic DNAmethylation changed showed a trend of first increasing and then decreasing. After rehydrating the P. ostii‘Fengdan’ plants treated with different degrees of drought,compared with the control,the total methylation rateof genomic DNA in P. ostii ‘Fengdan’ plants increased after rehydration. Under drought stress,there were4 variation patterns of DNA methylation in the genome of P. ostii ‘Fengdan’,which were represented by 15 bandtypes. Slight drought induced methylation at 39. 78% and demethylation at 40. 00% of the loci in the genome of P. ostii ‘Fengdan’;Severe drought induced methylation at 46. 00% and demethylation at 38. 60% of the loci inthe genome of P. ostii ‘Fengdan’,indicating that drought stress could induce changes in the methylation status of P. ostii ‘Fengdan’ genomic DNA.
