摘  要:试验旨在优化辣蓼黄酮的提取工艺。以辣蓼为试验材料,通过单因素试验考察液料比、乙醇含量、超声时间、超声温度对总黄酮含量的影响;确定影响总黄酮含量的主要因素和最佳范围后,以液料比、乙醇含量和超声时间为考察因素,以辣蓼黄酮含量为评价指标,通过响应面法优化辣蓼黄酮的提取工艺。结果显示,液料比、乙醇含量和超声时间是影响辣蓼黄酮含量的主要因素,当液料比20 mL/g、乙醇含量60%、超声时间40 min时,黄酮含量最高,分别是18.93%、18.07%和18.06%。液料比、乙醇含量和超声时间对辣蓼黄酮含量的影响大小依次是液料比>乙醇含量>超声时间,其中液料比和乙醇含量对辣蓼黄酮含量的影响达到极显著水平,超声时间不显著;液料比和超声时间之间交互作用对辣蓼黄酮含量影响最明显。验证试验结果显示,辣蓼黄酮最优提取工艺为液料比21 mL/g、乙醇含量62%、超声时间39 min,在此条件下黄酮的平均含量为19.108%。The aim of the experiment was to optimize the extraction technology of Polygonum hydropiper L.The effects of liquid-solid ratio,ethanol concentration,ultrasonic time and ultrasonic temperature on the content of total flavonoids were investigated by single factor test.After determining the main factors affecting the content of total flavonoids and the optimal range,the response surface method was used to optimize the extraction process of Polygonum hydropiper L.with the liquid-solid ratio,ethanol concentration and ultrasonic time as the evaluation index.The results showed that the liquidsolid ratio,the concentration of ethanol and the ultrasonic time were the main factors affecting the flavonoid content.When the liquid-solid ratio was 20 mL/g,the concentration of ethanol was 60%and the ultrasonic time was 40 min,the flavonoid content was the highest,which was 18.93%,18.07%and 18.06%,respectively.The effects of liquid-solid ratio,ethanol concentration and ultrasonic time on the flavonoid content were in the order of liquid-solid ratio>ethanol concentration>ultrasonic time,and the effects of liquid-solid ratio and ethanol concentration on the flavonoid content reached a very significant level,but the ultrasonic time was not significant.The interaction between the liquid-solid ratio and the ultrasonic time had the most obvious effect on the flavonoid content.The experimental results showed that the optimal extraction process was as follows liquid-solid ratio 21 mL/g,ethanol concentration 62%,ultrasonic time 39 min.Under these conditions,the average content of flavonoids was 19.108%.
