摘 要:为了有助于开展陆地棉PUP家族基因的生物学功能研究,也为后续陆地棉PUP基因在抗逆中功能的研究提供候选基因,利用生物信息学从陆地棉中鉴定PUP基因,并对PUP基因结构、保守结构域、顺式作用元件以及在胁迫条件下的表达等进行分析。结果表明,从陆地棉(TM-1)中共鉴定出31个PUP基因,依据进化分析结果,将这些基因分为3个亚支。染色体定位及基因复制事件分析结果表明,31个GhPUP家族基因分布在19条染色体上,且所有的PUP基因都由基因复制事件产生,复制类型为片段重复;基因结构的分析结果显示,陆地棉PUP基因大多数只有1个内含子,少数有2个或3个内含子,也有少数基因没有内含子;保守结构域分析表明,不同亚支上的PUP蛋白既有共同性也有特异性。顺式作用元件分析结果显示,GhPUPs基因启动子区域共有18个元件,这些元件分属于激素应答、胁迫/物理应答以及植物生长发育相关元件3类。同时,结合进化分析和转录组数据,筛选出了参与陆地棉胁迫应答的PUP基因。In order to facilitate studies on biological function of PUP family genes in Gossypium hirsutum,and provide candidate genes for subsequent studies on the function of PUP genes in stress resistance,in this study,31 PUP genes were identified in Gossypium hirsutum(TM-1)genome using bioinformatics,and could be classified into 3 subfamilies according to polygenetic analysis.The results of chromosome location and gene replication analysis showed that 31 GhPUP genes were distributed on 19 chromosomes,and all PUP genes were generated by gene replication events,and the duplication type was fragment duplication.Gene structure analysis indicated that most PUP genes had only one intron,a few had two or three introns,and a few had no introns.Conserved domain analysis showed that both commonality and specificity existed between PUP proteins in different subfamilies.Cis-acting elements analysis showed that there were 18 elements in the GhPUP genes promoter region,which were classified into three categories:hormone response,stress/physical response,and plant growth and development related elements.At the same time,combined with evolutionary analysis and transcriptome data,PUP genes involved in stress response in Gossypium hirsutum were screened.
