摘  要:本研究以包头市市花小丽花为外植体,探讨不同消毒时间、不同基础培养基、不同激素配比的培养基(启动培养基、增殖培养基、生根培养基)和炼苗移栽对小丽花组培苗的影响。结果表明,75%乙醇消毒30 s后,用0.1%HgCl2消毒10 min为最佳消毒时间,此时外植体的成活率最高,可达66.7%;MS+6-BA 1.0 mg/L+NAA0.2 mg/L为最佳的启动培养基,萌芽率达90.6%;MS+6-BA 0.4 mg/L+NAA 0.1 mg/L为最佳增殖培养基,增殖率为80.6%;MS+IBA 4 mg/L为最佳生根培养基,生根率为91.7%;炼苗后移栽到珍珠岩∶草炭∶园土=1∶1∶3的混合基质中,成活率可达90%。In this study,the city flower of Baotou City was used as the explant,the effects of different disinfection times,different basal medium,different hormone ratios of media(start-up medium,proliferation medium,rooting medium)and seedling refining and transplanting on tissue culture seedlings of Dahlia pinnata cv.were explored.The results showed that 75%alcohol disinfection for 30 seconds and 0.1%HgCl2 disinfection for 10 minutes were the optimal disinfection time,the survival rate of the explants was the highest,reaching 66.7%;MS+6-BA 1.0 mg/L+NAA 0.2 mg/L was the optimal start-up medium,with a germination rate of 90.6%;MS+6-BA 0.4 mg/L+NAA 0.1 mg/L was the optimal proliferation medium,with a proliferation rate of 80.6%;MS+IBA 4 mg/L was the optimal rooting medium,with a rooting rate of 91.7%;after refining,the seedlings was transplanted into the mixed substrate of perlite:peat:garden soil=1∶1∶3,and the survival rate reached 90%.
