摘 要:目的建立超高效液相色谱(UPLC)波长切换法同时测定芪参还五胶囊中羟基红花黄色素A、芍药苷、阿魏酸和毛蕊异黄酮葡萄糖苷4种化学成分含量。方法2021年5月至2022年1月,采用UPLC,色谱柱为WATERS ACQUITY UPLC BEH C18(2.1mm×50 mm,1.7μm),流动相为乙腈-0.2%甲酸水,梯度洗脱,流速0.3 mL/min,柱温35℃,进样量1μL,检测波长分别为检测波长为400 nm(羟基红花黄色素A)、230 nm(芍药苷)、320 nm(阿魏酸)和260 nm(毛蕊异黄酮葡萄糖苷)。结果羟基红花黄色素A、芍药苷、阿魏酸和毛蕊异黄酮葡萄糖苷的线性范围分别为1.34~26.80 mg/L(r=0.9998),2.46~49.20 mg/L(r=0.9999),0.582~11.64mg/L(r=0.9999),0.62~12.40 mg/L(r=0.9999);平均加样回收率分别为101.11%(RSD=1.64%)、99.62%(RSD=0.79%)、100.66%(RSD=2.13%)、101.93%(RSD=1.08%)。结论该方法简便、快速、准确性好,可为全面控制芪参还五胶囊的质量提供参考。Objective To establish an ultra performance liquid chromatography(UPLC)combined with switching wavelength method for simultaneous determination of four components in Qishen Huanwu capsules including hydroxysafflor yellow A,paeoniflorin,ferulic acid and calycosin-7-glucoside.Methods From May 2021 to January 2022,UPLC was performed on the column of WATERS ACQUITY UPLC BEH C18(2.1 mm×50 mm,1.7μm,),the mobile phase was acetonitrile-0.2%formic acid with gradient elution at the flow rate of 0.3 mL/min.The column temperature was 35℃,the injection volume was 1μL,and the detection wavelengths were 400 nm(hydroxysafflor yellow A),230 nm(paeoniflorin),320 nm(ferulic acid)and 260 nm(calycosin-7-glucoside).Results The linear ranges of hydroxysafflor yellow A,ferulic acid,paeoniflorin and calycosin-7-glucoside were 1.34~26.80 mg/L(r=0.9998),2.46~49.20 mg/L(r=0.9999),0.582~11.64 mg/L(r=0.9999),0.62~12.40 mg/L(r=0.9999),respectively.The average recoveries of the four components were 101.1%(RSD=1.6%),99.6%(RSD=0.79%),100.7%(RSD=2.1%),101.9%(RSD=1.1%),respectively.Conclusion The method is simple,accurate and reliable,which can be used for quality control of Qishen Huanwu capsules.
