摘 要:目的探讨不同剂量的地龙提取物对肺纤维化小鼠肺组织波形蛋白、α-平滑肌肌动蛋白表达水平及纤维化程度的影响。方法将90只C57雄性小鼠分为空白组、模型组、高剂量组、中剂量组和低剂量组,每组18只。除空白组外,其余4组通过腹腔注射百草枯建立肺纤维化模型。建模后1周,分别给予高剂量组、中剂量组、低剂量组小鼠灌服80 mg/(kg·d)、40 mg/(kg·d)、20 mg/(kg·d)的地龙提取物,给予空白组和模型组灌服等量的生理盐水,干预14 d。于建模后第14天、第21天和第28天,检测各组小鼠肺组织波形蛋白和α-平滑肌肌动蛋白表达水平;建模后第21天,采用HE染色法和Masson染色法分别观察小鼠肺组织纤维化及胶原沉积情况。结果建模后第14天、第21天及第28天,模型组小鼠肺组织的波形蛋白和α-平滑肌肌动蛋白表达水平均高于空白组(均P<0.05);建模后第21天、第28天,高剂量组、中剂量组、低剂量组小鼠肺组织的波形蛋白和α-平滑肌肌动蛋白表达水平均低于模型组(均P<0.05);建模后第28天,高剂量组小鼠肺组织的波形蛋白表达水平低于低剂量组,且α-平滑肌肌动蛋白表达水平均低于中剂量组与低剂量组(均P<0.05)。HE染色和Masson染色结果显示,模型组小鼠肺组织结构异常,正常肺泡结构消失,肺泡壁明显增厚,成纤维细胞增生明显,肺泡间隔和间质均可见到大量蓝色沉淀;相较于模型组,高剂量组、中剂量组、低剂量组小鼠肺部纤维化程度及胶原纤维沉淀程度明显减轻,肺泡结构相对完整,且高剂量组改善更明显。结论地龙提取物能有效抑制肺纤维化小鼠肺组织中波形蛋白和α-平滑肌肌动蛋白的表达,减少肺组织胶原沉积,延缓肺纤维化的进程,且高剂量[80 mg/(kg·d)]地龙提取物的改善效果更明显。Objective To explore the effect of Lumbricus extract with different doses on expressions of vimentin,α-smooth muscle actin and fibrosis degree in pulmonary tissues of mice with pulmonary fibrosis.Methods A total of 90 male C57 mice were assigned to blank group,model group,high-dose group,medium-dose group,or low-dose group,with 18 mice in each group.Except for the blank group,the pulmonary fibrosis model was established in the remaining 4 groups through intraperitoneal injection of paraquat.After 1 week of modeling,mice in the high-,medium-,and low-dose groups received administration of Lumbricus extract with 80 mg/(kg·d),40 mg/(kg·d),and 20 mg/(kg·d),respectively,and the blank and model groups received administration of normal saline with equivalent volume,for a 14-day intervention.On the 14 th,21 st,and 28 th day after modeling,the protein expressions of vimentin andα-smooth muscle actin in pulmonary tissues were detected in mice of various groups.On the 21 st day after modeling,fibrosis and collagen deposition in pulmonary tissues of mice were observed by using the HE and Masson staining,respectively.Results On the 14 th,21 st,and 28 th day after modeling,the model group exhibited higher protein expressions of vimentin andα-smooth muscle actin in pulmonary tissues as compared with the blank group(all P<0.05).On the 21 st,28 th day after modeling,the high-,medium-,and low-dose groups yielded lower protein expressions of vimentin andα-smooth muscle actin in pulmonary tissues as compared with the model group(all P<0.05).On the 28 th day after modeling,the high-dose group depicted a lower protein expression of vimentin as compared with the low-dose group,and a lower protein expression ofα-smooth muscle actin as compared with the medium-and low-dose groups(all P<0.05).The results of HE and Masson staining revealed that mice in the model group presented as abnormal pulmonary tissues structure,disappearance of normal alveolar structure,significantly thickened alveolar wall,significant fibroblast hyperplasia,p
