摘  要:目的探索仁青常觉通过干细胞因子(SCF)/细胞生长因子受体(c-kit)基因信号通路对胃动力低下大鼠的效应及机制。方法无特定病原体(SPF)级SD大鼠30只,用随机数字表法分为5组(每组6只):模型组;阳性对照(西沙必利0.4 mg/ml)组;仁青常觉低剂量(0.1 g/kg)组;仁青常觉中剂量(0.2 g/kg)组;仁青常觉高剂量(0.4 g/kg)组。建立胃动力低下大鼠模型,检测小肠推进率。酶联免疫吸附试验测定血液SCF的浓度;并对5-羟色胺(5-HT)和c-kit蛋白的表达进行免疫组织化学检测。结果与阳性对照组相比,模型组大鼠仁青常觉高剂量组大鼠小肠活动推进率、5-HT蛋白、血清中SCF含量及c-kit蛋白质均明显增高(P<0.01)。结论高剂量仁青常觉治疗可以缓解大鼠胃动力低下的情况。其机制可能与信号通路SCF/c-kit有关。Objective To investigate the mechanism underlying the effect of Rinchen Changjue(RCCJ),a Tibetan medicine,on rats with impaired gastric motility via the SCF/c-kit signaling pathway.Methods Thirty specific-pathogen-free(SPF)Sprague Dawley(SD)rats were randomized into 5 groups(n=6 each):blank model group,active control group(cisapride 0.4 mg/ml),low-dose(0.1 g/kg),medium-dose(0.2 g/kg),and high-dose RCCJ(0.4 g/kg)groups.Gastric hypomotility was modeled in rats.In all rats,we examined the small intestine propulsion rate.ELISA was used to measure the stem cell factor(SCF)level in blood.The expression of 5-HT and c-kit pro-tein was detected by immunohistochemistry.Results Compared with the other control groups,the intestinal propulsion rate,5-HT protein function,serum levels of SCF and c-kit protein were significantly higher in the rat models after administration of high-dose RCCJ(P<0.01).Conclusion Treatments with high-dose RCCJ alleviate gastric hypomotility in rats.The mechanism may be closely related to its regulation of the SCF/c-kit signaling pathway.
