摘 要:上皮细胞钠离子通道(ENaC)是广泛分布于全身各上皮细胞膜表面的离子通道,在呼吸道主要分布于肺泡上皮细胞和气管上皮细胞中,具有调节细胞内Na+浓度、维持体液平衡的功能。人类的ENaC由α、β、γ和δ4个亚单位构成,其中α-ENaC作为ENaC发挥功能的必需元件,是Na+转运的限速步骤。本文就近年来有关α-ENaC的结构、功能异常与呼吸系统相关疾病间的关系及潜在调节机制进行系统综述,并对不同药物的治疗效果进行总结,以期为揭示呼吸系统相关疾病的发病机制、促进新型药物的研发提供理论依据。The epithelial sodium channel(ENaC),widely distributed on the membrane of epithelial cells,is mainly expressed in alveolar and tracheal epithelial cells in respiratory system.ENaC plays an important role in regulating Na+concentration and maintaining homeostasis.ENaC is composed of four homologous subunits in human,namelyα,β,γ,andδ-ENaC,of whichα-ENaC is the necessary composition of ENaC playing its roles and the rate-limiting step for Na+transportation.In this review,we summarized the structure ofα-ENaC,the association betweenα-ENaC dysfunction and respiratory diseases,and the mechanism ofα-ENaC regulation.Finally,we discussed the therapeutic effects of various drugs.Hopefully it can provide theoretical basis for pathogenesis study of respiration related diseases and for exploring potential drugs for these diseases.
