基于功效关联的苍术质量标准研究

摘　　要：目的:针对现有苍术质量标准缺乏与功效关联的质量控制指标问题,探究“燥湿”功效的活性部位,基于功效关联的指标性成分建立质量标准。方法:采用大鼠水钠潴留模型,将雄性SD大鼠分为模型组、阳性药组、苍术总提物组、挥发油组和剩余部位组,造模后收集1~5 h的尿液并检测电解质离子浓度,考察苍术的利尿活性部位,进而以活性部位中的主要成分β-桉叶醇、苍术素和苍术酮为指标建立HPLC含量测定方法。结果:苍术的挥发油部位显著增加水钠潴留大鼠的尿量和尿液中Na+和Cl-的排出量,为苍术发挥利尿作用的有效部位。HPLC含量测定方法结果显示该方法线性关系良好,精密度、重复性、稳定性的RSD均小于3.0%,加样回收率为99.4%~101.9%。β-桉叶醇为茅苍术与北苍术的主要差异性成分,通过聚类分析可区分茅苍术和北苍术。结论:根据“燥湿”功效特点,结合利尿活性评价,确定苍术挥发油中的β-桉叶醇、苍术素和苍术酮为功效关联的质量控制指标,建立的含量测定方法可反映其有效性,且能反映茅苍术和北苍术的差异,对苍术的质量控制具有一定借鉴意义。Objective:In view of the lack of efficacy related index in current quality standards of Atractylodis rhizome(AR),the drying dampness effective fraction of AR was explored,and a quality standard based on efficacy related substances was established.Methods:SD male rats were selected for the normal saline load experiment,divided into model group,positive control group,AR total extract group,volatile oil group and residual part group.The rat urine samples within 1-5 h after modeling were collected for urinary electrolyte detection to investigate the diuretic effective fraction.Then an HPLC method was established for the determination ofβ-Eudesmol,atractylodin and atractylon,which were main components in the effective volatile oil fraction.Results:AR volatile oil was the diuretic effective fraction,which had good diuretic effects and increased the urine Na+and Cl-excretion significantly.The established HPLC method performed linear content determination with RSDs of precision,repeatability and stability less than 3.0%.The average recoveries were within the range of 99.4%-101.9%.β-Eudesmol was the differentiating component of Atractylodes lancea and A.chinensis.In the cluster analysis,A.lancea could be clearly distinguished from A.chinensis.Conclusion:According to the drying dampness efficacy and diuretic activity evaluation,β-Eudesmol,atractylodin and atractylon in volatile oil of AR are efficacy related index components.The established method for determination of these index components can reflect effectiveness of AR and the difference between A.lancea and A.chinensis,which has certain reference significance for the quality control of AR.
