名医验方聚精助育颗粒对无症状性弱精子不育症指纹图谱及功效关联物质的预测分析

摘　　要：目的 基于HPLC指纹图谱及网络药理学方法对聚精助育颗粒(JZG)进行健脾益肾、补血养精、活血通精功效关联物质预测分析。方法 基于前期JZG指纹图谱研究,依据可测性及可追溯性,对JZG药效活性成分进行筛选,并运用网络药理学方法构建出JZG“活性成分-靶点-通路”网络,进而预测出JZG功效关联物质。结果 根据前期JZG指纹图谱研究,有效标定共有峰29个,指认色谱峰6个,明确药效活性成分7个。分析JZG“活性成分-靶点-通路”关联网,可知关键靶点包括SRC、STAT3、JUN、PTPN11、ESR1等,关键通路包括癌症中蛋白多糖、PI3K-Akt信号通路、黏着斑、破骨细胞分化等,JZG的药效活性成分可能通过调节上述关键靶点及通路达到健脾益肾、补血养精、活血通精的作用。结论 本课题前期研究成功建立名医验方JZG的指纹图谱并对其色谱峰进行归属分析,进而对名医验方JZG的整体化及过程化进行质量控制;并通过网络药理学方法发现JZG药效活性成分具有传递性和溯源性的特点,同时与JZG的健脾益肾、补血养精、活血通精功效属性关系密不可分,可作为JZG潜在的功效关联物质,为JZG的进一步药效学研究提供理论依据。Objective To predict and analyze the related substances of Jujing Zhuyu Granules(JZG)on the basis of HPLC fingerprint and network pharmacology.Methods Based on the previous JZG fingerprint study,the active ingredients of JZG were screened according to the measurability and traceability,and the JZG"active ingredient target pathway"network was constructed by using network pharmacological methods,and then the JZG efficacy related substances were predicted.Results According to the previous JZG fingerprint study,29 peaks were effectively calibrated,6 chromatographic peaks were identified,and 7 active ingredients were identified.By analyzing JZG′s"active ingredient target pathway"correlation network,it can be seen that key targets include SRC,STAT3,JUN,PTPN11,ESR1,etc.Key pathways include proteoglycans in cancer,PI3K-Akt signal pathway,focal adhesion,osteoclast differentiation,etc.JZG′s pharmacodynamic activity component may achieve the role of strengthening spleen and kidney,enriching blood and nourishing essence,and promoting blood circulation and clearing essence by regulating the above key targets and pathways.Conclusions In the early stage of this research,the fingerprint of famous medical prescription JZG was successfully established and its chromatographic peaks were analyzed,so as to control the integrity and process of the famous medical prescription JZG;through the network pharmacological method,it was found that the active ingredients of JZG had the characteristics of transmissibility and traceability,and were closely related to JZG′s efficacy attributes of strengthening the spleen and kidney,nourishing blood and essence,and promoting blood circulation and essence circulation,which could be used as potential efficacy related substances of JZG,providing theoretical basis for further pharmacodynamic research of JZG.
