金龙消癌丸对非小细胞肺癌细胞体内外增殖凋亡作用的实验研究

摘　　要：目的探究金龙消癌丸对非小细胞肺癌(NSCLC)细胞体内外增殖凋亡作用。方法培养NSCLC细胞株A549、NCI-H1299,安替可胶囊、顺铂注射液做阳性对照组,金龙消癌丸做实验组。研究金龙消癌丸抑制NSCLC细胞体外增殖的效果,用CCK-8法检测;研究其促使NSCLC细胞体外凋亡的效果,采用流式细胞术。研究金龙消癌丸在体内效果,建立A549诱导裸小鼠皮下移植瘤模型,观察移植瘤体积、重量及抑瘤率。结果金龙消癌丸能有效抑制A549、NCI-H1299体外增殖,呈剂量依赖,IC50分别为4.268 mg/mL、9.911 mg/mL;有效促进A549、NCI-H1299细胞体外凋亡,细胞凋亡率分别达18.25%、10.82%;有效抑制NSCLC细胞体内增殖,高剂量优于低剂量,对肿瘤生长抑制率分别为35.30%和40.13%。结论金龙消癌丸能显著抑制NSCLC细胞体外增殖、诱导体外凋亡,能有效抑制体内NSCLC细胞移植瘤生长。Objective To explore the effect of Jinlong Xiaoai pill on the proliferation and apoptosis of non-small cell lung cancer(NSCLC)cells in vitro and in vivo.Methods NSCLC cell lines A549 and NCI-H1299 were cultured.Anticoc capsule and cisplatin injection were used as positive control group,and Jinlong Xiaoai pill was used as experimental group.The effect of Jinlong Xiaoai pill on inhibiting the proliferation of NSCLC cells in vitro was studied,and cell proliferation was detected by CCK-8 assay.Jinlong Xiaoai pill’s enhancing the apoptosis effect of NSCLC cells in vitro was studied,and cell apoptosis was detected by flow cytometry.When studying the effect of Jinlong Xiaoai pill on the proliferation and apoptosis of NSCLC cells in vivo,A549-induced subcutaneous xenograft tumor model in nude mice was established,weight and tumor inhibition rate of subcutaneously transplanted tumors in nude mice were observed.Results Jinlong Xiaoai pill could effectively inhibit the proliferation of A549 cells and NCI-H1299 cells in vitro,and the effect was dose-dependent.For the A549 cells,the IC50 was 4.268mg/ml,for the NCI-H1299 cells,IC50 was 9.911mg/ml.Jinlong Xiaoai pill could effectively promote the apoptosis of A549 cells and NCI-H1299 cells in vitro,and the effect was dose-dependent.Acting on A549 cells,the apoptosis rate was 18.25%,and NCI-1299 was 10.82%.Jinlong Xiaoai pill could effectively inhibit the proliferation of NSCLC cells in vivo,and the high dose was better than the low dose,the tumor growth inhibition rates were 35.30%and 40.13,respectively Conclusion Jinlong Xiaoai pills can significantly inhibit the proliferation of NSCLC cells in vitro and induce apoptosis in vitro,and can effectively inhibit the growth of NSCLC cells transplanted in vivo.
