摘 要:建立高效液相色谱法分析微拟球藻磷脂的主要组成与含量。从微拟球藻中提取出磷脂,经ZORBAX RX-SIL柱(4.6 mm×250 mm,5μm)分离,以甲醇-水(87∶13,V∶V)为流动相等度洗脱,流速为1 mL/min,柱温30℃,检测波长205 nm,外标法定量。磷脂酰胆碱、磷脂酰乙醇胺、磷脂酰肌醇、溶血磷脂酰胆碱在浓度0.1~1 mg/mL范围内线性关系良好,相关系数均大于0.996,加标回收率在85%~105%,相对标准偏差(RSD)小于3.1%。高效液相色谱法检测结果表明,微拟球藻磷脂的主要组成磷脂酰肌醇含量为19.32%,磷脂酰乙醇胺含量为18.74%,磷脂酰胆碱含量为11.86%,溶血磷脂酰胆碱含量为3.24%。该方法准确度高,重复性好,适用于微拟球藻中磷脂类化合物含量的测定。High-performance liquid chromatography(HPLC)was established to analyze the main composition and content of phospholipids in Nannochloropsis sp.Phospholipids were extracted from Nannochloropsis sp and separated on a ZORBAX RX-SIL column(4.6 mm×250 mm,5μm).With methanol-water(87:13,V∶V)as mobile phase,the flow rate was 1 mL/min,the column temperature was 30℃,the detection wavelength was 205 nm,and the external standard method was used.Phosphatidylcholine,phosphatidylethanolamine,phosphatidylinositol and lysophosphatidylcholine have a good linear relationship in the concentration range of 0.1~1 mg/mL.The correlation coefficients are all greater than 0.996.The recovery rate is 85%,and the relative standard deviation(RSD)is less than 3.1%.The results of high-performance liquid chromatography detection showed that the content of phosphatidylinositol,phosphatidylethanolamine,phosphatidylcholine and lysophosphatidylcholine were 19.32%,18.74%,11.86%and 3.24%,respectively.The method has high accuracy and good reproducibility,thereby is suitable for the determination of phospholipids in Nannochloropsis sp..
