猕猴桃DCL、RDR和AGO基因家族的鉴定与分析

摘  要:为研究猕猴桃RNAi抗病毒机制相关基因DCL、RDR和AGO。本研究采用blastp比对法和NPS@、SignalP-5.0及TMHMM 2.0等数据库对猕猴桃RNAi相关蛋白家族进行鉴定及其对应基础性质(如:亚细胞定位、二级结构等)的分析。结果在猕猴桃中共鉴定获得4个DCL、6个RDR和18个AGO基因,其中19个基因分布在13条染色体(即第3、6、9、12、13、16、17、18、19、23、26、27、29号染色体)上,另外9个基因未定位到染色体具体位置。AcAGO1d、AcAGO2a、AcAGO2b、AcAGO2d基因在19号染色体成簇存在。RDR基因和AGO2/7基因包含的内含子较少,其他的AGO和DCL基因包含的内含子较为丰富。这些猕猴桃基因均与番茄更近缘,它们编码的蛋白质含有不同比例的二级结构类型,无规则卷曲占50%左右,其次是α-螺旋和β-折叠。本研究完善了猕猴桃DCL、RDR和AGO编码基因的鉴定,后分析其对应的基础性质,以期为猕猴桃抗病毒研究奠定基础。Kiwifruit is facing many kinds of biological stress from virus in the production,which seriously threatens the safety of kiwifruit production.RNA interference(RNAi)is one of the important antiviral mechanisms in plants,three kinds of proteins were involved in RNAi:Dicer-like(DCL),RNA-dependent RNA Polymerase(RDR)and Argonaute(AGO).However,RNAi related genes in kiwifruit have not been identified by DCL,RDR and AGO.In this study,the blastp alignment method and databases such as NPS@,SignalP-5.0,and TMHMM 2.0 were used to identify the kiwifruit RNAi-related protein family and analyze their corresponding basic properties such as subcellular localization,secondary structure.The results showed that a total of 28 genes were found in kiwifruit,19 RNAi related genes were found on 13 chromosomes(chromosomes 3,6,9,12,13,16,17,18,19,23,26,27 and 29)and 9 genes were not placed on chromosomes.Among them,AcAGO1d,AcAGO2a,AcAGO2b and AcAGO2d genes existed in clusters on chromosome 19.In addition,RDR gene and part of AGO2/7 gene had few introns,while other AGO and DCL genes had abundant introns.In general,the kiwifruit genes are more closely related to the tomato,and the protein they encoded had different proportions of secondary structure,its random coil accounted for about 50%,followed by alpha helix and then extended strand.This study improved the identification of genes encoding DCL,RDR and AGO in kiwifruit,and their corresponding basic properties were analyzed,thus providing the research basis for kiwifruit antiviral research.
