摘 要:为明确甘肃省定西市一种新的赤芍根腐病病原菌及其生物学特性,采用常规组织分离法和孢子悬浮液离体接种法进行病原菌分离和致病性测定,基于形态学特征及内转录间隔区(Internal Transcribed Spacer, ITS)、核糖体大亚基(Ribosomal Large Subunit, LSU)和转录延伸因子1-α(Transcription elongation factor-1,TEF1-α)多基因序列分析进行病原菌鉴定,明确该病原菌为草莓垫壳孢(Coniella fragariae)。该病原菌生长最适温度为25℃,最适pH为6~7,全光照不利于菌丝生长和产孢,在PDA培养基上菌丝生长最快,在OA和黑麦培养基上产孢量最高,菌丝对乳糖和酵母膏的利用率最高。To clarify a new pathogen of root rot on Coniella fragariae and its biological characteristic in Dingxi of Gansu province,the pathogen was seperated and its pathogenicity was tested by the method of conventional tissue isolation and in vitro inoculation with spore suspension.The pathogenic fungus was identified to be C.fragariae based on morphology and ITS,LSU and TEF1-αpolygenic sequences.The optimal temperature for the growth of the pathogenic fungus is 25℃,the optimal pH value is 6-7,the full light is not conducive to hyphae growth and spore production,the hyphae growth is the fastest on PDA medium,the highest spore production on OA and rye medium,and the highest utilization rate of hyphae to lactose and yeast paste.
