摘 要:猪是人兽共患病病原戊型肝炎病毒(HEV)Ⅲ、IV基因型的重要贮存宿主。为了解云南省临沧市猪HEV的遗传进化情况,从规模猪场、猪散养户以及猪屠宰场,随机采集粪便153份、肝脏36份和胆汁54份,采用巢氏PCR进行HEV ORF2基因片段扩增、克隆、测序和生物信息学分析。结果显示:在243份样品中,检出137 bp目标片段阳性61份,阳性率为25.1%(61/243);对检出的阳性样品进行ORF2部分序列扩增,得到6份348 bp目标片段。6份348 bp目标片段间的核苷酸同源性为86.0%~95.6%,均为基因IV型;与HEVI-VⅢ型代表株的同源性分别为75.0%~81.7%、72.7%~77.0%、72.7%~79.3%、80.5%~94.1%、75.7%~80.7%、75.0%~81.0%、73.3%~80.3%和71.3%~76.1%,与我国人源代表毒株AJ272108BeijingHuman的同源性为81.3%~87.7%;基因亚型分析显示,6份348 bp目标片段中,除1份是4b亚型外,其他5份都属于4a亚型。结果表明,临沧市猪群中存在人兽共患病病原HEV基因Ⅳ型流行,且有通过肝脏、胆汁等产品感染传播的潜在风险,因此该地区应该重视猪HEV的公共卫生学意义。Pigs are the important reservoir of zoonotic hepatitis E virus(HEV)genotypes III and IV.In order to investigate the genetic evolution of HEV in pig populations in Lincang City,Yunnan Province,153 faecal samples,36 liver samples and 54 bile samples were randomly collected from large-scale farms,free-range households and slaughterhouses,then HEV ORF2 gene fragment amplification by nested PCR,cloning,sequencing and bioinformatic analysis were carried out.The results revealed that 61 positive samples containing targeted gene fragments with size of 137 bp were detected out,with a positive rate of 25.1%(61/243);partial sequence of ORF2 gene was amplified for the positive samples and 6 targeted fragments with size of 348 bp were obtained,whose nucleotide homology ranged from 86.0%to 95.6%and all belonged to genotype IV;their homologies were 75.0%-81.7%,72.7%-77.0%,72.7%-79.3%,80.5%-94.1%,75.7%-80.7%,75.0%-81.0%,73.3%-80.3%and 71.3%-76.1%to HEV representative strains type I to VIII,respectively,and 81.3%-87.7%to China human-origin representative strain of AJ272108BeijingHuman;analysis on genetic subtype showed that,5 out of 6 fragments with size of 348 bp fell into 4a subtype except for 1 that belonged to 4b subtype.In conclusion,zoonotic HEV genotype IV was prevalent in pig populations in Lincang City,with a potential risk of spreading via pig livers,bile and other products,against which,public health significance from porcine HEV should be concerned in the region.
