摘 要:正呼肠孤病毒(orthoreovirus)是一种分节段的双链RNA病毒,其宿主范围广泛,对人和动物健康构成了严重威胁。由于正呼肠孤病毒结构复杂,反向遗传系统构建困难,导致其相关研究滞后于其他RNA病毒。自2007年一种完全基于质粒的正呼肠孤病毒反向遗传系统被报道以来,关于正呼肠孤病毒基因和蛋白功能的研究不断增多。本文以哺乳动物正呼肠孤病毒(mammalian orthoreovirus,MRV)和禽正呼肠孤病毒(avian orthoreovirus,ARV)为例,概述了正呼肠孤病毒反向遗传系统和病毒载体研究的最新进展,并对未来发展趋势进行展望,以期为正呼肠孤病毒载体疫苗研发提供参考。Orthoreovirus is a segmented double-stranded RNA virus that occupies a wide host range and poses a serious threat to human and animal health.Due to its complex structure and the difficulty in constructing a reverse genetic system,relevant researches fall behind those on other RNA viruses.Since 2007 when an entirely plasmid-based reverse genetic system of orthoreovirus was reported,researches on the functions of orthoreovirus genes and proteins have been increasing.Taking mammalian orthoreovirus(MRV)and avian orthoreovirus(ARV)as examples,latest advances in the researches on reverse genetic system of orthoreovirus and its use as viral vectors were reviewed in the paper,and its development trend was prospected,with a view to providing a reference for future development of vector-based vaccines of orthoreovirus.
