摘 要:目的:观察参芪地黄汤化裁方对腺嘌呤慢性肾衰大鼠血清肌酐(SCr)、尿素氮(BUN)及肾脏病理改变的影响。方法:取Wistar大鼠60只,随机分为6组,分别为正常组、模型组、阳性组和参芪地黄汤化裁方低、中、高剂量组,每组10只;以腺嘌呤造模,检测各组大鼠血清SCr、BUN含量,进行组间比较分析。采用HE染色、Masson染色观察肾脏病理改变。结果:与正常组比较,模型组血清SCr、BUN含量显著升高(P<0.01);与模型组比较,阳性组、参芪地黄汤化裁方3剂量组SCr、BUN含量均有显著下降(P<0.05、P<0.01);HE染色显示模型组肾小球数量减少,囊腔扩张,肾小管扩张,管腔内可见黄色沉积物;Masson染色显示肾组织纤维化,结构显色区分不清。各给药组上述病理表现较模型组轻,其中阳性组和高剂量组病理改变减轻更明显。结论:参芪地黄汤化裁方能显著延缓腺嘌呤致慢性肾衰大鼠肾功的下降,并改善其肾组织病理变化。Objective:To observe the effects of Modified Shenqi Dihuang Decoction on serum creatinine(SCr),urea nitrogen(BUN),and renal pathological changes in rats with chronic renal failure caused by adenine.Methods:60 Wistar rats were randomly divided into 6 groups:normal group,model group,positive drug group,and low,medium,and high dose groups of Modified Shenqi Dihuang Decoction,with 10 rats in each group;Using adenine as a model,the levels of SCr and BUN in each group of rats were measured for comparative analysis between groups.HE staining and Masson staining were used to observe renal pathological changes.Results:Compared with the normal group,the serum levels of SCr and BUN in the model group were significantly increased(P<0.01);Compared with the model group,there were significant differences(P<0.05,P<0.01)in the content of SCr and BUN between the positive drug group and the three dose groups with Modified Shenqi Dihuang Decoction;HE staining showed a decrease in the number of glomeruli,dilation of the capsule and renal tubules,and visible yellow deposits in the lumen in the model group;Masson staining showed renal ti-ssue fibrosis,with unclear structural color differentiation.The above pathological manifestations in each medication group were lighter than those in the model group,with the positive group and high-dose group showing more significant reduction in pathological changes.Conclusion:Modified Shenqi Dihuang Decoction can significantly delay the decline of renal function in rats with chronic renal failure caused by adenine,and improve the pathological changes of renal tissue.
