摘  要:目的:探究苍耳子含药血清对免疫球蛋白E(IgE)致敏的肥大细胞脱颗粒的抑制作用和调控途径。方法:小鼠喂服苍耳子水溶液,制备含药血清;MTT法检测不同浓度苍耳子含药血清对肥大细胞活性的影响。IgE致敏肥大细胞,实验分为正常对照组、模型对照组、10%苍耳子含药血清组和富马酸酮替芬组。药物干预后,乳酸脱氢酶(LDH)法和甲苯胺蓝染色检测细胞毒性和脱颗粒率;ELISA检测细胞炎症活性介质β氨基己糖苷酶A(β-hexosaminidase A)、白三烯C4(LTC4)和组胺(histamine)的释放情况;F-actin微丝染色观察细胞形态变化。结果:苍耳子含药血清对正常肥大细胞活性无明显影响(P>0.05)。与正常对照细胞相比,模型对照组细胞毒性和脱颗粒率增加,β氨基己糖苷酶A、白三烯C4、组胺的释放显著增加,细胞变形数量显著增加(P<0.01)。与模型对照组相比,10%苍耳子含药血清和富马酸酮替芬组细胞毒性和脱颗粒率显著降低,β氨基己糖苷酶A、白三烯C4、组胺的释放显著下降,细胞变形数量显著减少(P<0.01)。结论:苍耳子含药血清可抑制肥大细胞变形、脱颗粒,并减少β氨基己糖苷酶A、白三烯C4、组胺的释放。Objective:To explore the inhibitory effect and regulatory pathway of containing Xanthium serum(cXs)on degranulation of IgE sensitized mast cell.Methods:Mice fed with Xanthium aqueous solution to prepare cXs.The effect of cXs on the activity of mast cells was detected by MTT test.IgE sensitized mast cells,and cells were divided into normal control group,model control group,10%cXs group and Ketotifen fumarate group.After intervention,LDH test and toloniumchloride staining were used to detect the toxicity and degranulation rate of cells;ELISA was used to detect the release ofβ-hexosaminidase,leukotriene C4(LTC-4)and histamine of cells;F-actin microfilaments was used to observe morphological changes of cells.Results:cXs had no significant effect on activity of mast cells(P>0.05).Compared with normal control group,the cytotoxicity and degranulation rate of cells of model control group were significantly increased,the release ofβ-hexo-saminidase,LTC-4,histamine,and the number of cell deformations were significantly increased(P<0.01).Compared with model control group,the cytotoxicity and degranulation rate of cells of 10%cXs and Ketotifen fumarate groups were significantly reduced,the release ofβ-hexosaminidase,LTC-4 and histamine was significantly reduced;and the number of cell deformation was significantly reduced(P<0.01).Conclusion:The cXs can inhibit deformation and degranulation of IgE sensitized mast cell,and reduced the release ofβ-hexosaminidase,LTC-4 and histamine.
