摘 要:目的:探讨地龙提取物对肺炎支原体肺炎小鼠肺损伤相关修复因子增殖细胞核抗原(PCNA)及细胞外调节激酶(ERK)的表达影响。方法:采用20μL(1×10^(6)CCU)肺炎支原体菌液滴鼻3 d建立肺炎支原体肺炎模型。将60只BALB/c小鼠随机分为正常组,模型组,地龙高(4.7 g/kg)、中(2.35 g/kg)、低剂量组(1.175 g/kg),阳性对照组(阿奇霉素22.5 mg/kg),每组10只;各组小鼠给药10 d后,采用HE染色法、PCR方法及免疫组化法观察各组小鼠肺组织病理变化及肺组织PCNA、ERK的mRNA及ERK蛋白表达。结果:HE染色结果显示,与模型组相比,地龙中、高剂量组均能改善肺组织病变,减少炎性细胞浸润,肺组织PCNA、ERK的mRNA和ERK蛋白表达水平较模型组显著升高(均P<0.05)。结论:地龙可通过影响PCNA、ERK的mRNA及蛋白表达水平修复肺炎支原体肺炎小鼠的肺上皮组织。Objective:To invstigate the effects of Earthworm Extract on the expressions of PCNA and ERK related to Mycoplasma pneumoniae pneumonia(MPP).Methods:The MPP models were established by the nasal instillation of 20μL 1×10^(6) CCU MP.60 BALC/c mice were divided into normal control group,model group,positive control group(Azithromycin 22.5 mg/kg),and Earthworm high(4.7 g/kg),medium(2.35 g/kg)and low group(1.175 g/kg)dose groups,each group contained 10 mice.They were given the corresponding medicine for 10 days,the pathological changes in the lung ti-ssues of mice were observed by HE staining,the expressions of PCNA and ERK mRNA and ERK protein in lung epithelial cells were determined by RT-PCR and immunohistochemistry.Results:The HE staining results showed that compared with the model group,both the medium and high dose groups of Earthworm could improve lung tissue lesions,reduce inflammatory cell infiltration,and significantly increase the expression levels of PCNA,ERK mRNA,and ERK protein in lung tissue(all P<0.05).Conclusion:Earthworm could repair the pulmonary epithelial tissue of Mycoplasma pneumoniae pneumonia by inhibiting the expressions of PCNA and ERK.
