摘 要:目的:探讨蛹虫草多糖对高脂高胆固醇诱导ob/ob小鼠肝组织脂质代谢、炎症反应和肠道菌群的影响。方法:40只雄性ob/ob小鼠随机分为模型组、非诺贝特组[20 mg/(kg·d)]及蛹虫草多糖低、中、高剂量组[25、50、100 mg/(kg·d)],均以高脂高胆固醇饮食(脂肪34.9%,胆固醇22.8%)饲养。12周末,取新鲜肝组织匀浆,ELISA方法检测肝组织脂质代谢相关酶脂肪酸合成酶(FAS)、长链脂酰辅酶A脱氢酶(LCAD)活性及C反应蛋白(CRP)、单核细胞趋化蛋白1(MCP-1)、血清淀粉样蛋白A(SAA)、肿瘤坏死因子α(TNF-α)、核转录因子κB亚基p65(NF-κB p65)、白介素6(IL-6)含量。另取小鼠新鲜粪便,采用16S rDNA扩增子测序,检测多糖对小鼠肠道菌群的影响。结果:与模型组相比,高剂量蛹虫草多糖可显著降低FAS含量(P<0.05),显著提升LCAD含量(P<0.01);高、中剂量蛹虫草多糖显著抑制CRP表达(P<0.01,P<0.05),同时明显降低MCP-1和SAA表达(P<0.05)。与模型组比较,3个剂量蛹虫草多糖能显著抑制NF-κB p65和TNF-α表达(P<0.01,P<0.05),高剂量多糖可降低IL-6水平(P<0.05),而低、中剂量组无显著性差异。16S rDNA检测显示,高剂量多糖可逆转肠道有益菌群Marinifilaceae、Odoribacter、Lachnospiraceae等在模型组小鼠中的丰度降低。结论:泰山蛹虫草多糖可通过调控肠道菌群改善肝组织脂质代谢,减轻炎症反应。Objective:To explore the effects of Cordyceps Militaris Polysaccharide from Taishan o n hepatic lipids metobolism,inflammation response and intestinal floras in ob/ob mice which induced by high-fat and high-choleeterol diet.Methods:40 male ob/ob mice were randomly divided into model group,Fenofibrate group and three doses Polysaccharide groups.Mice were all given high-fat and high-cholesterol diet.The contents of FAS and LCAD in liver were detected by ELISA method.The levels of CRP,MCP-1 and SAA in liver were also determined with ELISA method.In the meanwhile,the contents of IL-6,NF-κBp65 and TNF-αin liver were detected with the same methods.The intestinal floras in gut was tested by 16 SrDNA methods.Results:In contrast to the models,the activities of FAS was inhibited by the high dose Cordyceps Militaris Polysa-ccharide,and the activities of LCAD were raised significantly(P<0.01).The levels of CRP,MCP-1 and SAA were decreased by the high dose and the middle dose Cordyceps Militaris Polysaccharide.In contrast to the models,the three doses Cordyceps Militaris Polysaccharide declined the levels of TNF-α,NF-κBp65 and IL-6(P<0.01,P<0.05).Conclusion:The Cordyceps Militaris Polysaccharide from Taishan could improving lipid metabolism and inflammatory response in liver by regulating intestinal floras.
