摘 要:传统的体外细胞检测技术多数建立在光学基础上,操作复杂且会对细胞造成一定程度的损伤,很难保证在细胞不被破坏的条件下检测药物对细胞的影响。为解决此难题,通过有限元仿真软件COMSOL仿真并确定了直径5μm的微柱阵列电极作为细胞基底,采用细胞阻抗传感技术监测了不同浓度的紫杉醇溶液对人结肠癌细胞(SW480)黏附生长的影响。实验表明,随着细胞培养液中紫杉醇溶液浓度升高,人结肠癌细胞在圆柱基底上的阻抗不断减小,很好的从阻抗角度反映了药物对细胞的损伤程度,具有很好的应用价值和发展空间。Most of the traditional in vitro cell detection technologies are based on optics,whose operation is complex and will cause a certain degree of damage to cells.It is difficult to ensure that the effects of drugs on cells are detected without destroyed.In order to solve this problem,the finite element simulation software COMSOL was used to simulate and determine the micro column(diameter:5μm)array electrode as the cell substrate.The cell impedance sensing technology was used to monitor the effect of different concentrations paclitaxel solution on the adhesion and growth of human colon cancer cells(SW480).The experiment shows that the impedance of human colon cancer cells on the column base decreases with the increase of paclitaxel concentration,which reflects the influence of drugs on cells from the perspective of impedance,and has good application value and development space.
