摘　　要：采用全长转录组文库结合三代高通量测序的方法得到远海梭子蟹Sox9基因的全长序列,通过引物设计及PCR产物直接测序验证三代测序结果,最终准确地获得Sox9基因序列。对Sox9基因全长序列进行生物信息学及基因进化分析。结果表明该基因CDS基因序列全长1461bp,蛋白质的无规则卷曲、α-螺旋、β-折叠区域分别占58.64%、25.93%、0%。进化树分析结果表明,远海梭子蟹Sox9与三疣梭子蟹的Sox10基因同源性较高,其核酸序列和氨基酸序列的相似性分别为96.24%和98.77%。远海梭子蟹的Sox9基因鉴定和蛋白质结构预测与功能分析,将为该物种遗传资源挖掘与利用分析奠定重要基础。Combined with three-generation high-throughput sequencing,the full-length transcriptome library was used to obtain the full-length sequence of the Sox9 gene of Portunus pelagicus,and the final accurate sequence of the Sox9 gene was obtained through primer design,PCR amplification and validation of the three-generation sequencing results.Bio-informatics analysis of the full-length sequence of the resulting Sox9 gene was performed to determine the evolutionary status of the gene and the species.The results showed that the full length of Sox9 gene CDS was 1461bp,and the irregularly coiled,α-helical andβ-folded regions of the protein accounted for 58.64%,25.93%and 0%respectively.The evolutionary tree analysis showed that the Sox9 gene of Portunus pelagicus was highly homologous to the Sox10 gene of P.trituberculatus,with 96.24%similarity and up to 98.77%amino acid sequence comparison.The sequence validation and protein structure prediction of the Sox9 gene of Portunus pelagicus will be useful for the conservation of germplasmresources and genetic analysis of the species.
