摘  要:目的探究长链非编码RNA(lncRNA)在疟原虫感染过程中表达谱变化特征。方法建立P.y 17XL感染BALB/c鼠疟模型,计数红细胞感染率,记录生存率;使用RNA-seq方法确认P.y 17XL感染的BALB/c小鼠lncRNA的差异表达谱。GO和KEGG分析以阐明lncRNAs在疟疾感染过程中对免疫功能的调节作用。结果与正常对照组小鼠相比,P.y 17XL感染BALB/c小鼠脾脏中lncRNAs表达谱发生明显改变,132个lncRNAs表达上调,159个lncRNAs表达下调。通过GO和KEGG分析发现,明显差异表达的lncRNAs包括3个已知和9个新发现的优势lncRNAs,即ENSMUST00000146154.1、ENSMUST00000181191.1、ENSMUST00000202081.1、LNC_000258、LNC_000871、LNC_000962、LNC_002093、LNC_004354、LNC_006284、LNC_004518、LNC_004350和LNC_003730均与疟疾免疫应答密切相关。结论P.y 17XL感染改变了宿主lncRNAs的表达谱,宿主-寄生虫相互作用通过lncRNAs调控了抗疟疾免疫应答。本研究为进一步研究疟原虫与宿主细胞之间作用机制提供理论依据。This study explored the characteristics of long non-coding RNA(lncRNA)expression profiles in the process of malaria infection.BALB/c mice were infected with P.y 17XL,and parasitemia and survival rates were monitored.RNA-seq was used to confirm the differential expression of lncRNAs in P.y 17XL-infected BALB/c mice.GO and KEGG analysis were performed to elucidate the regulatory roles of lncRNAs in immune function in Malaria.Analysis of significant changes in lncRNA expression revealed 132 up-regulated and 159 down-regulated lncRNAs.GO and KEGG analysis indicated that the clearly differentially expressed lncRNA included three known and nine novel lncRNAs(ENSMUST00000146154.1,ENSMUST00000181191.1,ENSMUST00000202081.1,LNC_000258,LNC_000871,LNC_000962,LNC_002093,LNC_004354,LNC_006284,LNC_004518,LNC_004350 and LNC_003730)that correlated with the immune response in malaria.P.y 17XL infection altered the host lncRNA expression profile.Host-parasite interaction regulates anti-malaria immunity via lncRNAs.This study is expected to provide a theoretical basis for further research on the interaction mechanism between Plasmodium and host cells.
