摘 要:目的为了解云南省大理州亚洲带绦虫的遗传多样性。方法本研究采集到大理(DL)、巍山(WS)、弥渡(MD)、漾濞(YB)和洱源(EY)5个地区的131份亚洲带绦虫样本,基于线粒体12S rRNA基因进行扩增,运用MEGA7.0和DNASP5.10.01软件对所得序列进行分析,计算其碱基含量、遗传多样性参数、遗传分化系数和基因流以及遗传距离等数据。结果5个地区的亚洲带绦虫12S rRNA基因片段大小均为493 bp,平均A、T、G、C的碱基含量为28.3%、43.2%、19.4%和9.1%;共定义76个单倍体型,其中单倍型多样性为0.9830,核酸多样性为0.0332;遗传分化指数和基因流范围分别为-0.01913~0.04080和5.87745~22.49795;遗传距离为0.0023~0.0229,大理与巍山之间的遗传距离最大,漾濞与洱源群体之间的遗传距离最小。结论大理州5个地区亚洲带绦虫的线粒体12S rRNA基因的遗传多样性水平较高,基因交流频繁,但种群间的遗传分化较弱,本研究为亚洲带绦虫遗传多样性提供了基础数据。In order to understand the genetic diversity of Taenia asiatica in Dali Prefecture,Yunnan Province.In this study,131 samples of T.asiatica were collected from Dali(DL),Weishan(WS),Mitu(MD),Yangbi(YB)and Eryuan(EY)of Dali Prefecture,Yunnan province.12S rRNA gene of mitochondrial were amplificated.MEGA7.0 and DNASP5.10.01 software were performed to analyze the obtained sequences,the data of base content,genetic diversity parameters,genetic differentiation coefficient,gene flow and genetic distance were calculated.The results showd that the 12S rRNA gene fragments of T.asiatica samples from 5 regions were 493 bp in size from each sample,the average contents of A,T,G and C were 28.3%,43.2%,19.4%and 9.1%,respectively;76 haplotypes were defined,the haplotype diversity was 0.9830 and the nucleic acid diversity was 0.0332.The range of genetic differentiation index and gene flow were-0.01913~0.04080 and 5.87745~22.49795,respectively;The genetic distance is 0.0023~0.0229,the genetic distance between DL and WS is the largest,and the genetic distance between YB and EY is the smallest.The mitochondrial 12S rRNA gene of T.asiatica in five regions of Dali Prefecture has a high level of genetic diversity and frequent gene exchange,but the genetic differentiation among populations is weak.The results of this study provided basic data for the genetic diversity of T.asiatica.
