摘 要:目的探究白及抗炎药效部位(EFBS)对骨关节炎的潜在治疗作用及其可能分子机理。方法通过UPLC-QTF-MS技术对EFBS主要化学组成进行鉴定;采用IL-1β诱导的软骨组织炎症模型和ADTC5软骨细胞炎症模型,通过测定炎症因子水平,胶原基质降解情况和Ⅱ型胶原蛋白α1(COL2A1)和基质金属蛋白酶13(MMP13)基因表达水平,综合分析EFBS对骨关节炎防治作用和机制。结果共鉴定了EFBS中12个主要峰,其中菲类和联苄类各6个。EFBS可剂量依赖性抑制IL-1β诱导的软骨组织IL-6、MCP-1升高和胶原基质降解(P<0.05),其中40μg/mL处理时对IL-6和MCP-1的抑制率分别达88.736%±19.305%和69.189%±26.366%。同样,EFBS也可浓度依赖性抑制IL-1β诱导的ADTC5软骨细胞胶原基质降解(P<0.05),其可能与促进COL2A1基因表达和抑制MMP13基因表达有关。结论EFBS主要由菲类和联苄类组成,其对IL-1β诱导软骨和软骨细胞炎症反应和基质降解具有较好抑制作用,展现出潜在的防治骨关节炎作用,值得进一步深入研究。Objective To explore the potential therapeutic effects and possible molecular mechanisms of the anti-inflammatory active fraction(EFBS)of Bletilla striata on osteoarthritis.Methods The main chemical composition of EFBS identified using UPLC-QTF-MS technology.IL-1βinduced cartilage tissue inflammation model and ADTC5 chondrocyte inflammation model were carried out to evaluate the preventive effect and mechanism of EFBS on osteoarthritis,by measuring the levels of inflammatory factors,collagen matrix degradation,collagen type II alpha 1(COL2A1)and matrix metalloproteinase 13(MMP13)gene expression levels.Results A total of 12 main peaks in EFBS were identified,including 6 phenanthrenes and 6 benzenes compounds.EFBS could dose-dependently inhibit IL-1βinduced elevation of IL-6 and MCP-1 in cartilage tissue and degradation of collagen matrix(P<0.05).When treated with 40μg/mL of EFBS,its inhibitory rates on IL-6 and MCP-1 reached 88.736%±19.305%and 69.189%±26.366%,respectively.Similarly,EFBS could also inhibit IL-1βinduced collagen matrix degradation in ADTC5 chondrocytes in a concentration dependent manner(P<0.05),which might be related to promoting COL2A1 gene expression and inhibiting MMP13 gene expression.Conclusion EFBS is mainly composed of phenanthrene and benzyl compounds,which have a good inhibitory effect on IL-1βinduced inflammatory response and matrix degradation in cartilage and chondrocytes,demonstrating potential preventive and therapeutic effects on osteoarthritis,and is worthy of further in-depth research.
