摘  要:目的 分析2019年潍坊市一株登革病毒(Weifang 2019)全基因组序列及关键性氨基酸位点的突变情况。方法 收集登革病毒感染病例的血清样本,对初检阳性样本进行病毒分离、全基因组测序,构建进化树,对该序列进行核苷酸/氨基酸相似性分析。结果 本研究中,分别以登革病毒1型(DENV-1)参考毒株(NC_001477.1)和原型株(EU848545.1)为对照,Weifang 2019核苷酸基因组的全序列分别检出94个和95个氨基酸位点突变,其中有57个突变位点一致。其中,3个结构蛋白和7个非结构蛋白均存在突变,蛋白NS5、E和NS1位点突变占比前3位。核苷酸序列和氨基酸序列分析发现Weifang2019与Guangdong 2014(99.5%/99.3%)、Wenzhou 2014(99.6%/99.4%)、Guangdong 2016(99.4%/99.2%)、Henan 2014(99.5%/99.3%)处于同一进化分支,且相似性均大于99%。结论 2019年潍坊地区一株登革病毒Weifang 2019与Guangdong 2014(MN018287.1)、Wenzhou 2014(KR024708.1)、Guangdong 2016(MN018286.1)和Henan 2014(KP686070.1)的遗传距离较近、相似性较高,提示Weifang 2019可能由上述地区毒株遗传进化而来。Objective To analyze the sequence of the complete genome a dengue virus strain from Weifang City in 2019(Weifang 2019)and the mutation in key amino acid sites.Methods Serum samples of dengue virus infection cases were collected,and virus isolation and whole genome sequencing were performed on the initial positive samples,so as to construct evolutionary tree for analysis and amino acid/nucleotide similarity analysis.Results In this study,taking the reference strain of DENV-1(NC_001477.1)and the prototype strain(EU848545.1)as control,94 and 95 amino acid mutations were detected in the complete nucleotide genome in Weifang 2019,of which 57 mutations were identical.Among them,mutations were found in 3 structural proteins and 7 nonstructural proteins,and mutations at NS5,E and NS1 accounted for the first three spots.Nucleotide sequence and amino acid sequence analysis showed that Weifang 2019 and Guangdong 2014(99.5%/99.3%),Wenzhou 2014(99.6%/99.4%),Guangdong 2016(99.4%/99.2%),Henan 2014(99.5%/99.3%)were in the same evolutionary clade,with similarity above 99%.Conclusion In 2019,a dengue virus Weifang 2019 showed relatively close genetic distance and high similarity with Guangdong 2014(MN018287.1),Wenzhou 2014(KR024708.1),Guangdong 2016(MN018286.1)and Henan 2014(KP686070.1),which suggested that Weifang 2019 might genetically evolve from the strains of the above area.
