摘 要:目的研究柳茶水提物(SAW)对大鼠离体主动脉血管条张力的调节作用及其作用机制。方法制作大鼠离体主动脉血管条,固定于灌流肌槽中,观察不同浓度SAW对大鼠离体主动脉血管条张力的影响及对去甲肾上腺素(NA)、氯化钙(CaCl_(2))、氯化钾(KCl)诱导离体大鼠血管条的影响,以确定SAW的血管活性;同时研究阿托品、普萘洛尔、吲哚美辛、L-精氨酸(L-ARG)、维拉帕米等阻断剂对SAW扩张NA预收缩血管作用的影响,以确定SAW舒张血管的作用机制。结果随着SAW质量浓度的升高,大鼠离体血管条张力逐渐降低;SAW对NA及CaCl_(2)诱导的血管收缩有明显抑制作用,对KCl诱导的血管收缩作用不明显;阿托品、吲哚美辛、L-ARG可显著抑制SAW扩张NA预收缩血管的作用,普萘洛尔、维拉帕米对SAW扩张NA预收缩血管作用的影响不明显。结论SAW可通过多种途径使得血管平滑肌张力降低,舒张血管,从而降低血压,此作用与受体操控Ca^(2+)通道、M受体、前列腺素类物质、一氧化氮相关,与β受体、L-型Ca^(2+)通道无关。Objective To study the regulating effect of Sibiraea angustata water extract(SAW)on the tension of isolated vascular strip in rats and its mechanism.Methods Isolated aorta strips in rats were prepared and fixed in the perfusion tank.The effects of different concentrations of SAW on the tension of isolated aorta strip in rats and the tension of isolated vascular strip in rats induced by noradrenalin(NA),calcium chloride(CaCl_(2))and potassium chloride(KCl)were observed to determine the vascular activity of SAW.At the same time,the effects of atropine,propranolol,indometacin,L-arginine(L-ARG),verapamil and other blockers on SAW dilated pre-constriction blood vessels induced by NA were studied to determine the vasodilation mechanism of SAW.Results With the increase of SAW mass concentration,the tension of isolated aorta strips in rats decreased gradually.SAW has significant inhibitory effect on the vasoconstriction induced by NA and CaCl_(2),but hasn’t significant inhibitory effect on the vasoconstriction induced by KCl.Atropine,indometacin and L-ARG could significantly inhibit the effect of SAW on dilating pre-constriction blood vessels induced by NA,while propranolol and verapamil had no significant effect.Conclusion SAW can reduce vascular smooth muscle tension,dilate the blood vessels and reduce blood pressure through various ways.This effect is related to receptor manipulating Ca^(2+)channels,M receptors,prostaglandins and nitric oxide,and is not related toβreceptors and L-type Ca^(2+)channels.
