摘 要:目的:观察针刺枕视皮质对应区对青光眼模型大鼠视神经的影响,分析可能机制。方法:将100只SD大鼠中的80只构建青光眼模型,高眼压状态持续4周后,随机分为模型组、联合针刺组、常规针刺组、枕视皮质组;剩余20只为空白组。联合针刺组针刺球后穴、风池穴、太阳穴、行间穴、枕视皮质对应区,常规针刺组针刺球后穴、风池穴、太阳穴、行间穴,枕视皮质组针刺枕视皮质对应区。每2日针刺1次,连续针刺3周。空白组、模型组不针刺。观察各组视网膜神经节细胞(RGC)数量及形态变化,免疫组织化学法检测人超大B细胞淋巴瘤因子(Bcl-XL)、信号传导及转录激活因子3(stat3)光密度值(OD),免疫印迹法检测Bcl-XL、stat3蛋白表达。结果:与模型组比较,联合针刺组、枕视皮质组、常规针刺组RGC数量均升高(P<0.05),且联合针刺组RGC数量高于常规针刺组及枕视皮质组(P<0.05)。空白组RGC形态大致正常,排列整齐、紧密;模型组RGC形态异常,有拉长变形,甚至出现溶解情况,排列无序;联合针刺组RGC形态较正常,排列较整齐,无明显溶解、变形;枕视皮质组和常规针刺组RGC形态相对正常,排列较模型组整齐,RGC轻度变形,无明显溶解情况。与空白组比较,各组模型大鼠Bcl-XL、stat3 OD均升高(P<0.05),且联合针刺组、枕视皮质组、常规针刺组Bcl-XL、stat3 OD均高于模型组(P<0.05),联合针刺组Bcl-XL、stat3 OD均高于枕视皮质组及常规针刺组(P<0.05)。与空白组比较,各组模型大鼠Bcl-XL、stat3蛋白表达均升高(P<0.05),且联合针刺组、枕视皮质组、常规针刺组Bcl-XL、stat3蛋白表达均高于模型组(P<0.05),联合针刺组Bcl-XL、stat3蛋白表达均高于枕视皮质组及常规针刺组(P<0.05)。结论:针刺枕视皮质对应区联合常规穴位可以激活青光眼模型大鼠视神经保护通路,保护高眼压下的视神经,修复被高眼压损伤的视神经及RGC,改善视神经�Objective:To observe the effect of acupuncture at the corresponding area of occipital visual cortex on optic nerve of glaucoma model rats and analyze the possible mechanism.Methods:Total 80 out of 100 SD rats were used to establish glaucoma models,and after a four-week period of high intraocular pressure,they were randomly divided into the model group,the combined acupuncture group,the conventional acupuncture group,and the occipital visual cortex group.The remaining 20 rats were the blank group.The combined acupuncture group was treated with acupuncture at the acupoints of Qiuhou(EX-HN7),Fengchi(G20),Taiyang(EX-HN5),Xingjian(LR2)and the corresponding area of the occipital visual cortex.The conventional acupuncture group was treated with acupuncture at the acupoints of Qiuhou,Fengchi,Taiyang,Xingjian.The occipital visual cortex group was treated with acupuncture at the corresponding area of the occipital visual cortex.Needling was performed once every 2 days for 3 consecutive weeks.No acupuncture was performed in the blank group and model group.The number and morphological changes of retinal ganglion cell(RGC)in each group was detected.The optical density(OD)of human large B-cell lymphoma factor(Bcl-XL)and signal transduction and transcriptional activation factor 3(stat3)were detected by immunohistochemistry.The expressions of Bcl-XL and stat3 were detected by immunohistochemistry.Results:Compared with the model group,the number of RGC in the combined acupuncture group,occipital visual cortex group,and conventional acupuncture group increased(P<0.05);And the number of RGC in the combined acupuncture group was higher than that in the conventional acupuncture group and the occipital visual cortex group(P<0.05).The morphology of RGC in the blank group was generally normal,with orderly and compact arrangement;The RGC morphology in the model group was abnormal,showed up as elongated and deformed,and even dissolved,with disordered arrangement;The morphology of RGC in the combined acupuncture group was relatively normal
