摘 要:目的:优化人参茎叶中人参皂甙含量检测前处理的最佳工艺,确定人参茎叶原料中的人参皂甙含量检测方法并进行方法学考察。方法:采用正交实验优化人参茎叶中人参皂甙含量检测前处理工艺,采用高效液相色谱法测定人参总皂甙含量,并通过线性关系、精密度试验、重复性试验、稳定性试验、加样回收试验考察此方法的科学性与可行性。结果:原料前处理的最佳工艺为氧化镁加入量0.10 g,过40目筛,在50℃条件下超声提取1.5 h;方法精密度、重复性、稳定性良好,平均加样回收率为99.4%。结论:本文确立了人参茎叶原料的最优前处理方法,为人参皂甙含量的检测提供参考。Objective:To optimize the pretreatment technology of ginsenoside in stem and leaf of Panax ginseng,determine the detection method for stem and leaves of Panax ginseng raw materials,and conduct methodological studies.Methods:The optimal pre-treatment process for the detection of ginsenoside content in stem and leaves of Panax ginseng is optimized using orthogonal experiments.The total ginsenoside content is determined by highperformance liquid chromatography,and the scientific and feasibility of this method is investigated through linear relationship,precision test,repeatability test,stability test,and sample recovery test.Results:The optimal pretreatment process of raw materials is to add 0.10 g of magnesium oxide,pass through a 40 mesh sieve,and extract it by ultrasound at 50 ℃ for 1.5 hours.The method has good precision,repeatability,and stability,with an average spiked recovery rate of 99.4%.Conclusion:The pretreatment method of stem and leaves of Panax ginseng raw materials is established to provide reference for the detection method of ginsenoside content.
