摘　　要：目的:建立一种应用于片状载体培养的Vero细胞计数方法。方法:Vero细胞于细胞瓶内培养,分别于24 h、48 h、72 h、96 h和120 h取样检测上清液中葡萄糖含量,同时消化细胞并计数。以葡萄糖累计消耗量为X轴,以细胞收获数量为Y轴,获得葡萄糖累计消耗量和细胞数量的关系方程,从而建立一种细胞计数的方法。通过计算反应器中细胞生长消耗的葡萄糖量来计算细胞数量,从而优化控制细胞生长。结果:得到葡萄糖消耗量与细胞数量线性方程为Y=4.107×10~8X;Vero细胞在反应器内培养6 d,累计消耗葡萄糖71.28 g,所获得细胞数量为2.93×10^(10)个。结论:建立了葡萄糖消耗量与细胞数量的线性关系,即每消耗1 g葡萄糖对应的细胞数量为4.107×10~8个,可作为片状载体细胞培养过程中细胞计数的参考。Objective:To establish a Vero cell counting method applied to the culture of flake carriers.Methods:Vero cells are cultured in cell flasks,and samples are taken at 24 h,48 h,72 h,96 h,and 120 h to detect the glucose content in the supernatant,and the cells are digested and counted.Taking the cumulative glucose consumption as the X axis and the number of cells harvested as the Y axis,the relationship equation between the cumulative glucose consumption and the number of cells is obtained,so as to establish a method for cell counting.Optimizing cell growth is controlled by calculating the cell number by calculating the amount of glucose consumed for cell growth in the reactor.Results:The linear equation of glucose consumption and cell number is Y=4.107×10~8X;Vero cells are cultured in the reactor for 6 days,the cumulative glucose consumption is 71.28 g,and the number of cells obtained was 2.93×10~(10).Conclusion:A linear relationship between glucose consumption and cell number is established.That is,the number of cells corresponding to each consumption of 1 g of glucose is 4.107×10~8,which can be used as a reference for cell count in the flake carrier cell culture process.
