摘 要:目的分析脐带血中DLK1和GRB10基因甲基化水平与正常出生体质量婴幼儿生长发育的相关性,探讨影响婴幼儿生长发育的原因和机制。方法以舟山市孕妇队列中符合研究标准的100名孕妇及其子代作为研究对象,其中,50例生长发育迟缓婴幼儿为迟缓组,50例生长发育正常婴幼儿为对照组。检测脐带血中DLK1和GRB10基因的甲基化水平,并在婴幼儿第6个月、12个月、18个月和24个月时评价婴幼儿的生长发育情况。采用Wilcoxon秩和检验分析子代生长发育迟缓组与正常组的甲基化水平分布差异;采用线性回归模型分析脐带血DLK1、GRB10基因甲基化水平与婴幼儿身高和体质量的相关性。结果Wilcoxon秩和检验结果显示:迟缓组与正常组婴幼儿脐带血DLK1和GRB10基因不同片段甲基化水平比较,差异均无统计学意义(P>0.05)。整合DLK1和GRB10基因CG、CHG和CHH类型各片段后,迟缓组DLK1基因CG、CHG和CHH位点甲基化水平分别为37.0(35.1,39.1)、98.6(98.3,99.1)和98.3(97.9,98.6),正常组分别为36.3(33.6,38.4)、98.6(98.2,99.0)和98.2(97.9,98.5),两组之间差异均无统计学意义(P>0.05)。迟缓组GRB10基因CG、CHG和CHH位点甲基化水平分别为1.7(1.5,1.9)、72.5(64.7,82.5)和75.4(71.7,80.1),正常组分别为1.5(1.4,1.8)、74.2(66.7,78.9)和76.0(71.5,80.8),两组之间差异均无统计学意义(P>0.05)。调整新生儿性别和喂养方式等混杂因素后,线性回归模型分析结果显示,脐带血DLK1和GRB10基因甲基化水平与婴幼儿第6个月、12个月、18个月和24个月龄身高、体质量均无统计学关联(P>0.05)。结论脐带血DLK1和GRB10基因甲基化水平与正常出生体质量婴幼儿的生长发育没有关联。Objective To analyze the correlation between the methylation levels of DLK1,GRB10 genes in cord blood and the growth and development of infants,and to explore the possible mechanisms of growth retardation in infant.Methods 100 pregnant women and their offspring who met the research criteria in Zhoushan pregnant women cohort were taken as the research objects,of which 50 infants with growth retardation were the retardation group,and 50 infants with normal growth and development were the control group.To detect the methylation level of DLK1 and GRB10 genes in cord blood,and evaluate the growth and development of infants at the 6th,12th,18th and 24th months.The wilcoxon rank sum test was used to analyze the difference of methylation level distribution between the growth retardation group and the normal group;Linear regression model was used to analyze the correlation between the methylation level of DLK1 and GRB10 genes in cord blood and the height and body mass of infants.Results The wilcoxon rank sum test showed that there was no statistical difference in the methylation levels of different segments of DLK1,GRB10 genes in cord blood between the growth retardation group and the normal group(P>0.05).After integration of CG,CHG and CHH type fragments of DLK1,GRB10 genes,The methylation levels of CG,CHG and CHH sites of DLK1 gene in delayed group were 37.0(35.1,39.1),98.6(98.3,99.1)and 98.3(97.9,98.6),respectively.the normal groups were 36.3(33.6,38.4),98.6(98.2,99.0)and 98.2(97.9,98.5),there was no significant difference between the two groups(P>0.05).The methylation levels of CG,CHG and CHH sites of GRB10 gene in the delayed group were 1.7(1.5,1.9),72.5(64.7,82.5)and 75.4(71.7,80.1),respectively.the normal groups were 1.5(1.4,1.8),74.2(66.7,78.9)and 76.0(71.5,80.8),there was no statistical significance between the two groups(P>0.05).After adjusting for the confounding factors such as newborn gender and feeding mode,the linear regression model analysis results showed that the methylation levels of DLK1,GRB10 genes in
