基于Wnt/β-catenin信号通路探讨四君子汤对胃癌脾气虚证裸鼠的作用机制

摘　　要：目的:探究四君子汤对胃癌脾气虚证裸鼠的分子学作用机制。方法:将20只BALB/c雄性裸鼠随机分为正常组6只和模型组14只,正常组常规喂养,模型组以番泻叶药液灌胃结合饥饱失常法建立脾气虚证模型,再行胃癌原位移植手术造模,造模成功的14只裸鼠随机抽取7只按10.92 g/kg灌胃四君子汤(四君子汤组),剩余7只裸鼠仍为模型组。记录各组裸鼠精神状态,皮毛色泽,反应力、活动度等行为学表征及肿瘤质量与抑瘤率;采用免疫印迹法和qPCR法检测肿瘤组织Wnt/β-catenin的蛋白及其mRNA相对表达量。结果:正常组裸鼠活动正常,皮毛柔顺有光泽,大便正常;模型组裸鼠精神萎靡不振,毛发杂乱,反应迟钝,活动度欠佳,弓背懒动,肛门湿润红肿,大便不成形,呈溏状;四君子汤组裸鼠上述情况均有改善。与模型组相比,四君子汤组裸鼠肿瘤质量减轻(P<0.05),抑瘤率为47%,Wnt1基因、β-连环蛋白、MYC癌基因和基质金属蛋白酶7的mRNA及其蛋白表达降低(P<0.05),糖原合酶激酶3β的mRNA及其蛋白表达升高(P<0.05)。结论:四君子汤可能是通过调控Wnt/β-catenin信号通路来改善胃癌对机体的影响。Objective:To survey the molecular mechanism of Sijunzi Tang in the treatment of gastric cancer of spleen Qi deficiency pattern in nude mice.Methods:Twenty BALB/c male nude mice were randomized into six ones in the normal group and 14 in the model group,the normal group accepted normal feeding,and the model group adopted intragastric administration with Fanxieye soup and the method of hunger and aberration to establish the model of spleen Qi deficiency pattern,and then perform in-situ transplantation of gastric cancer for modeling,seven nude mice were randomly chosen from 14 ones after successful modeling and they were drenched with Sijunzi Tang(Sijunzi Tang group),and the remaining seven mice were selected as the model group.To record mental state,the color and lustre of skin and hair,behavioral characterization including reaction and activity,the weight of tumor and tumor-inhibition rate of the nude mice in different groups;Western blotting method and qPCR method were used to detect the relative expressions of Wnt/β-catenin protein and its mRNA in tumor tissue.Results:Nude mice in the normal group had normal activity,with smooth and shiny coat,and normal stools;the ones in the model group were depressed,with disorganized fur,slow response,poor activity,bow-backed and slouching,wet and red anus,unformed and loose stool;the above conditions of the nude mice in Sijunzi Tang group were improved.Compared with the model group,the weight of tumor reduced in nude mice in Sijunzi Tang group(P<0.05),the tumor-inhibition rate was 47%,the expressions of Wnt1 gene,β-Collagen,MYC cancer gene and MMP-7 mRNA and protein lowered(P<0.05),the levels of glycogen synthase kinase 3βmRNA and protein increased(P<0.05).Conclusion:Sijunzi Tang could improve the influence of gastric cancer on human body possibly through regulating of Wnt/β-catenin signaling pathway.
