摘 要:目的构建亚砷酸钠(SA)与环磷酰胺(CTX)联合染毒模型,探究SA与CTX对雄性小鼠生殖及肝功能的联合毒性。方法将40只成年健康雄性昆明小鼠以体重作为分组因素,按完全随机分组设计分为SA组、CTX组、SA+CTX组(联合组)、对照组,采用腹腔注射法染毒,共持续5周。结果体重:对照组和SA组小鼠体重呈时间-效应关系增长。小鼠染毒期间,SA组体重变化与对照组相比差异无统计学意义;CTX组(P<0.05)与联合组(P<0.05)小鼠体重呈波动下降趋势。生殖系统:与对照组相比,CTX组(P<0.05)与联合组(P<0.05)小鼠睾丸脏器系数均明显降低。联合组精子密度、精子总数介于单染毒实验组之间。苏木精-伊红(HE)染色结果显示,CTX组小鼠睾丸内生精细胞数量减少、生精小管管径减小。肝:与对照组相比,CTX组的血清谷丙转氨酶(ALT)(P<0.05)和谷草转氨酶(AST)(P<0.05)明显升高,联合组与CTX组相比,ALT(P<0.05)和AST(P<0.05)降低;与对照组相比,联合组肝超氧化物歧化酶(SOD)活力升高(P<0.05),CTX组肝丙二醛(MDA)含量增加(P<0.05);相比于CTX组,联合组肝MDA降低(P<0.05)。结论SA与CTX均可对小鼠造成生殖损伤和肝损伤,而SA和CTX共同染毒则可能具有拮抗作用。Objective To establish a combined poisoning model for sodium arsenite(SA)and cyclophosphamide(CTX)to investigate into the impact of the joint toxicity of SA and CTX on the reproductive function and liver function of male mice.Methods Forty sexually mature and healthy KM male mice were divided into four groups according to completely random block design using body weight as the block factor:SA group,CTX group,combination group and control group.The mice in each group were intraperitoneally injected with poison for five weeks.Results Effects on body weight:Both the control group and the SA group underwent an increase in body weight,which was time-dependent.During the exposure,the weight change of the single SA group was not statistically significant when compared with the control group.However,the mice in the CTX group(P<0.05)and combined group(P<0.05)showed a fluctuating decline in weight.Effects on the reproductive system:The testicular organ coefficient decreased significantly in both the CTX(P<0.05)and the combined group(P<0.05).The sperm density and total sperm count of the combined group were between those of the individual poisoning groups.Hematoxylin-eosinstaining(HE)results revealed a decrease in spermatogenic cells and seminiferous tubule diameter in the CTX group.Effects on the liver:The levels of alanine aminotransferase(ALT)(P<0.05)and aspartate aminotransferase(AST)(P<0.05)were significantly increased in the CTX group,while there was no statistically significant difference between the combined and the control group.Compared with the control group,liver superoxide dismutase(SOD)activity decreased(P<0.05)and malondialdehyde(MDA)content increased(P<0.05).Conclusion It was found that both SA and CTX could cause reproductive and liver damage.When SA and CTX were combined,they exhibited an antagonistic effect.
