摘 要:筛选对冷杉梢斑螟高致病性线虫,并探究昆虫线虫对冷杉梢斑螟体内营养物质变化及保护酶和解毒酶的影响。通过控制变量法从5个品系昆虫病原线虫中筛选具有高致病性的病原线虫,同时采用吸光光度法研究了其对冷杉梢斑螟体内主要营养物质及保护酶和解毒酶活性,观察随着处理时间的延长,体内主要营养物质及酶活性的变化。从5个昆虫线虫品系发现小卷蛾斯氏线虫AⅡ品系对冷杉梢斑螟4龄幼虫的防治效果显著,处理120 h后对冷杉梢斑螟的最高校正死亡率达到91.94%。幼虫体内营养物质含量均围绕对照组营养物质含量呈现波动变化。幼虫体内总糖质量分数,在处理后随侵染时间增加明显升高,12 h达到最大,随后迅速下降,在24 h降至最低25.62 mg·g^(-1),与对照组差异显著(P<0.05);脂肪质量分数随着时间增加变化趋势与总糖质量分数变化趋势相似,处理12 h和36 h达到最高值,且显著高于对照组;蛋白质质量分数随着时间的增加呈上升趋势,12 h达到最大值4.28 mg·g^(-1)且与对照间有显著差异(P<0.05),随后下降,至18 h之后呈缓慢上升趋势,至48 h时,低至2.27 mg·g^(-1),并显著低于对照(P<0.05);多酚氧化酶(PPO)、超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、乙酰胆碱酯酶(AchE)、谷胱甘肽S-转移酶(GST)均呈现先增加后减少的趋势,PPO呈现先减少后增加趋势。侵染8 h时,CAT表达量达到最高19.3μmol·min^(-1)·g^(-1);18 h时,虫体内SOD、Ache和GST均达到峰值,分别为721.00、45.40和1133.68 nmol·min^(-1)·g^(-1);24 h时,SOD和PPO表达量降至最低值,48 h时GST降至最低635.38 nmol·min^(-1)·g^(-1),PPO达到最高值624.20 nmol·min^(-1)·g^(-1)。昆虫病原线虫的侵入破坏了寄主的防御系统,影响寄主体内营养物质与保护酶、解毒酶的含量,可为冷杉梢斑螟的生物防治及昆虫病原线虫制剂的开发和应用提供理论依据。The experiment was conducted to screen Entomopathogenic Nematodes(EPNs)that is highly pathogenic to Dioryctriaabietella,and explore the nutrient changes of D.abietella and the influence of EPNs on D.abietella’s protective enzymes and detoxification enzymes.The highly pathogenic EPN was screened from 5 strains of EPNs by the control variable method.At the same time,the absorbance spectrophotometry was used to study the activities of the main nutrients,protective enzymes and detoxification enzymes of EPN on D.abietella,and the changes of the activities of the main nutrients and enzymes were observed with the extension of the treatment time.From 5 EPNs,it was found that the fourth instar larvae of D.abietella were significantly controlled by Steinernemacarpocapsae(Weiser)AⅡ,and the highest corrected mortality rate of the fir stem borer reached 91.94%after 120-h treatment.The content of nutrients in the larvae fluctuated around the content of nutrients in the control group.The total carbohydrate content with the infection time after treatment,reaching the maximum at 12 h,then decreased rapidly,and decreased to the lowest 25.62 mg/g at 24 h,which was significantly different from the control group(P<0.05).The change trend of free fatty acid content with time was similar to that of total sugar content,reaching the highest value at 12 and 36 h after treatment,which was significantly higher than that of the control group.The protein content increased with time,reaching the maximum value of 4.28 mg/g at 12 h,which was significantly different from that of the control(P<0.05),and then decreased.After 18 h,it slowly increased,and at 48 h it was as low as 2.27 mg/g,which was significantly lower than that of the control(P<0.05).SOD,CAT,AchE and GET all showed a trend of increasing first and then decreasing,while PPO showed a trend of decreasing first and then increasing.At 8-h infection,CAT expression reached the highest level of 19.3μmol/min/g.At 18 h,SOD,Ache and GST in the worm reached their peak values,which were 721,4
