摘 要:目的:验证丹参酚酸B(SalB)通过上调细胞内活性氧(ROS)含量抑制LOVO细胞侵袭与迁移以及SalB对LOVO细胞侵袭和迁移的量效相关性调控。方法:体外培养人大肠癌LOVO细胞,并将其分成6组,Control组、SalB-L组、SalB-M组、SalB-H组、H_(2)O_(2)组和SalB-M+NAC组,进行分组干预。显微镜下观察LOVO细胞形态,采用流式细胞术测定细胞内ROS浓度,Transwell法检测细胞侵袭和迁移能力。结果:显微镜下可见,SalB干预后LOVO细胞生长抑制、细胞伪足变短或消失;低浓度SalB对LOVO细胞内ROS含量无影响(P>0.05),中、高浓度SalB浓度依赖性促进细胞内ROS生成(P<0.01),中浓度SalB诱导产生的ROS可被ROS清除剂NAC完全清除(P<0.01);低浓度SalB对LOVO细胞侵袭、迁移能力无影响(P>0.05),中、高浓度SalB可浓度依赖性抑制LOVO细胞侵袭、迁移(P<0.01),中浓度SalB对细胞侵袭、迁移的抑制作用可被NAC完全逆转(P<0.01)。结论:SalB可通过上调细胞内ROS浓度量效相关性抑制大肠癌LOVO细胞侵袭和迁移。Objective:To verify that Salvianolic acid B(SalB)inhibits the invasion and migration of LOVO cells by up-regulating the content of intracellular reactive oxygen species(ROS),and to verify the dose-dependent regulation of SalB on the invasion and migration of LOVO cells.Methods:Human colorectal cancer LOVO cells were cultured in vitro and were divided into Control group,SalB-L group,SalB-M group,SalB-H group,H_(2)O_(2) group and SalB-M+NAC group.Interventions were carried out according to the different groups.The morphology of LOVO cells were observed under microscope,flow cytometry was uesd to detect intracellular ROS concentration and Transwell test was used to detect the invasion and migration of cells.Results:Microscopically,the growth of LOVO cells were inhibited and the pseudopodia of LOVO cells became shorter or disappeared after SalB intervention.Low concentration of SalB had no effect on the content of ROS in LOVO cells(P>0.05),medium and high concentrations of SalB promoted the production of ROS in the cells in a concentration-dependent manner(P<0.01),and ROS induced by medium concentration of SalB could be completely cleared by ROS scavenger NAC(P<0.01);Low concentration of SalB had no effect on the invasion and migration of LOVO cells(P>0.05),medium and high concentrations of SalB could inhibit the invasion and migration of LOVO cells in a concentration-dependent manner(P<0.01),and the inhibitory effect of medium concentration of SalB on the cell invasion and migration could be completely reversed by NAC(P<0.01).Conclusion:SalB can inhibit the invasion and migration of colorectal cancer LOVO cells by up-regulating intracellular ROS concentration in a dose-dependent manner.
