摘 要:为了探究生长激素释放激素(growth hormone releasing hormone,ghrh)基因对梭鲈(Sander lucioperca)生长调控的作用,本研究利用RACE技术克隆获得了ghrh cDNA基因全长序列,并利用实时荧光定量PCR技术对其组织表达模式和大小个体间表达模式进行了分析。结果显示:ghrh基因cDNA全长为1 100 bp,包含494 bp的5′非编码区、180 bp的3′非编码区和编码141个氨基酸的开放阅读框。氨基酸多重比对发现,梭鲈ghrh与河鲈(Perca fluviatilis)、黄金鲈(P.flavescens)的ghrh氨基酸序列相似性分别为95.04%和94.33%,ghrh基因在鱼类和哺乳动物中都存在一个GLUCA保守结构域(65~91 aa)。ghrh基因在梭鲈各组织均有表达,脑组织中相对表达量最高,垂体次之,与其他组织相比差异显著。ghrh基因表达量与梭鲈体质量呈正相关,脑、肝和肌肉组织中ghrh基因表达量均为体质量极大组高于极小组,其中,脑组织中差异显著。因此,ghrh基因可以作为梭鲈生长候选基因用于分子选择育种。The full-length sequence of ghrh cDNA gene was cloned by RACE technology in this study to reveal the growth hormone releasing hormone(ghrh)gene of Sander lucioperca and explore its sequence charac-teristics and the relationship between its expression and growth traits.Meanwhile,this study also provided a theoretical basis of the molecular mechanism of its growth.The results showed that the full-length cDNA of ghrh gene was 1100 bp,including 494 bp 5′untranslated region,180 bp 3′untranslated region and open reading frame encoding 141 amino acids.Multiple amino acid alignments indicated that the similarity between ghrh gene of S.lucioperca,that of Perca fluviatilis(95.04%)and P.flavescens(94.33%)was the highest.ghrh gene had a GLUCA conservation domain(65~91 aa)in fish and mammals.The gene expression level of ghrh gene in different tissues and individuals of extreme body mass size was analyzed by real-time fluorescence quantitative PCR technology.ghrh gene was expressed in all tissues of S.lucioperca,with the highest relative expression in the brain,followed by the pituitary,and the relative expression in the two tissues was significantly different from other tissues.The amount of expression in skin and muscle was similar,but the difference was insignificant.The expression of ghrh gene was positively correlated with the body mass of S.lucioperca.The expression levels of ghrh gene in the brain,liver and muscle tissues of the big group were higher than that of the small group.Significant difference results showed that no significant difference was found in brain,liver and muscle tissues.Therefore,ghrh gene can be used as molecular selective breeding of a growth candidate gene of S.lucioperca.This study provides a reference for further understanding the regulation mechanism of growth axis genes on the growth and development of S.lucioperca.
