摘 要:[目的]研究含龙井提取物和薄荷脑等成分的中药草本液的体外抗菌性能及抗菌机理。[方法]选用大肠杆菌(Escherichia coli)、金黄色葡萄球菌(Staphylococcus aureus)和白色念珠菌(Candida albicans)检测草本液的最小抑菌浓度(MIC)、最低杀菌浓度(MBC)、抑菌环,利用Live-Dead试剂盒染色确定草本液抗菌活性,在扫描电子显微镜下观察草本液作用后细菌表面微观结构变化并分析作用机理。[结果]MIC和MBC结果表明草本液在稀释2倍之后对大肠杆菌、金黄色葡萄球菌和白色念珠菌仍具有抑制作用和杀菌作用;抑菌率、抑菌环与Live-Dead荧光染色表明草本液有显著抗菌作用;草本液作用后的细菌表面结构发生明显变化。[结论]草本液对大肠杆菌、金黄色葡萄球菌和白色念珠菌具有显著的抗菌效果。[Objective]To study the antibacterial effect and antibacterial mechanism of a traditional Chinese medicine herbal liquid that contains Longjing extract and menthol in vitro.[Method]E.coli,S.aureus and C.albicans were selected to determine the antibacterial rate and antibacterial activity by detecting minimum inhibitory concentration(MIC),minimum bactericidal concentration(MBC),antibacterial ring diameter and Live-Dead fluorescence staining.The effect of traditional Chinese medicine herbal liquid on the surface structure was observed by scanning electron microscope(SEM)and analyzed its antibacterial mechanism.[Result]The result of MIC and MBC showed that the herbal liquid diluted two times had inhibitory and bactericidal effects on E.coli,S.aureus and C.albicans.Antibacterial rate,antibacterial ring and Live-Dead fluorescence staining revealed that the herb liquid had significant antibacterial effect.The surface structure of bacteria changed obviously.[Conclusion]The herb liquid has significant antibacterial effect on E.coli,S.aureus and C.albicans.
