摘 要:目的探究冬凌草甲素对多发性骨髓瘤(MM)U266细胞、RPMI8226细胞增殖和凋亡的影响。方法通过细胞培养和处理方法,采用0、10、20、30、40、50μmol/L的冬凌草甲素处理MM相关U266细胞、RPMI8226细胞48 h。采用MTT法测定U266细胞、RPMI8226细胞的增殖情况,采用Annexin V-FITC/PI染色试剂盒检测U266细胞、RPMI8226细胞凋亡情况,并采用蛋白质印迹法测定Bax、Bcl-2蛋白表达。结果处理48 h后,随着冬凌草甲素浓度(10~50μmol/L)的增加,U266细胞、RPMI8226细胞的细胞增殖抑制率、细胞凋亡率均显著升高,差异均有统计学意义(P<0.05)。处理48 h后,随着冬凌草甲素浓度(10~50μmol/L)的升高,促凋亡蛋白Bax表达量逐渐升高,而抗凋亡蛋白Bcl-2表达量逐渐降低,差异均有统计学意义(P<0.05)。结论冬凌草甲素具有抑制MM细胞增殖和诱导细胞凋亡的潜力,其作用机制可能与促进Bax表达,抑制Bcl-2表达,改变细胞Bcl-2/Bax比例,调节细胞凋亡有关。Objective To investigate the effect of dongling grass methylene on the proliferation and apoptosis of multiple myeloma(MM)U266 cells,RPMI8226 cells.Methods The cell culture and treatment methods was used,MM related U266 cells and RPMI8226 cells were treated with 0,10,20,30,40,and 50μmol/L oridonin for 48 hours.The proliferation of U266 cells and RPMI8226 cells was measured by MTT method,the apoptosis of U266 cells and RPMI8226 cells was detected by Annexin V-FITC/PI staining kit,and the expression of Bax and Bcl-2 proteins was determined by Western blotting.Results After 48 hours of intervention,as the concentration of oridonin increases,the cell proliferation inhibition rate and apoptosis rate of U266 cells and RPMI8226 cells were significantly increased,and the differences were statistically significant(P<0.05).After 48 hours of intervention,as the concentration of oridonin A increased,the expression of pro apoptotic protein Bax gradually were significantly increased,while the expression of anti apoptotic protein Bcl-2 gradually were significantly decreased,and the differences were statistically significant(P<0.05).Conclusion The results of this study indicate that dongling grass methylene has the potential to inhibit proliferation and induce apoptosis in multiple myeloma cells.This provides a scientific basis for the use of dongling grass methylin as a candidate drug for the treatment of multiple myeloma.
