摘  要:目的探究榆苋方保留灌肠对大肠湿热型溃疡性结肠炎(UC)大鼠模型Pxr、NF-κB及炎症因子和结肠黏膜的影响。方法选取Wista大鼠(SPF级)成年大鼠53只,体质量200~220 g,扬州大学动物实验中心提供,雌雄各半,适应性饲养1周后建立UC大鼠模型,随机分为6组:模型组、空白对照组、低剂量治疗组、中剂量治疗组、高剂量治疗组、阳性对照组等,空白对照组8只,余每组9只。空白对照组不建模,其余组别建立溃疡性结肠炎模型,模型组不使用干预治疗药物,低剂量治疗组、中剂量治疗组、高剂量治疗组分别使用榆苋方不同剂量药物保留灌肠,阳性对照组使用柳氮磺吡啶灌肠。治疗14 d后观察结肠黏膜损伤情况,结肠黏膜中白细胞介素(IL)-10、干扰素(INF)-γ水平变化,以及Pxr mRNA、NF-κB p65 mRNA、IκB-αmRNA表达情况。结果与空白对照组相比,UC大鼠模型组、SASP组、榆苋方各剂量组INF-γ水平显著升高,IL-10水平明显降低(P<0.05);经榆苋方治疗后,随着给药剂量逐渐增高,INF-γ水平明显降低(P<0.05),IL-10水平明显升高(P<0.05);高剂量榆苋方组IL-10、INF-γ水平与SASP组阳性对照组相比差异无统计学意义(P>0.05)。模型组与正常对照组比较,Pxr和IκB-αmRNA表达水平显著降低,SASP阳性对照组和榆苋方各剂量组给药干预治疗后,Pxr和IκB-αmRNA表达明显增加;模型组与空白对照组进行比较,模型组NF-κB p65 mRNA表达明显增加,经给药干预治疗后,其表达水平有显著下降,高剂量榆苋方组Pxr mRNA、NF-κB p65 mRNA、IκB-αmRNA水平与SASP组阳性对照组相比差异无统计学意义(P>0.05)。结论榆苋方保留灌肠对大肠湿热型溃疡性结肠炎大鼠具有肠黏膜损伤修复及炎症因子调控作用。Objective To investigate the effect of Yuxian Decoction(榆苋方)preserved enema on Pxr,NF-κB,inflammatory factors and colon mucosa in rat models of large intestine damp heat type ulcerative colitis(UC).Methods 53 Wista rats(SPF level)adult rats with a weight of 200-220 g were selected,provided by the animal experiment center of Yangzhou university,male and female,and a UC rat model was established after 1 week of adaptive rearing,which were randomly divided into six groups:model group,blank control group,low dose treatment group,medium dose treatment group,high dose treatment group,positive control group,9 cases in each group,except for the blank control group.The blank control group did not model,the remaining groups established ulcerative colitis models,the model group did not use intervention drugs,the low-dose treatment group,the medium-dose treatment group,and the high-dose treatment group used different doses of Yuxian Decoction to retain the enema,and the positive control group used sulfasalazine enemas.After 14 days of treatment,to observe the lesions of the colonic mucosa,the levels of interleukin-10(IL-10),interferon-γ(INF-γ)in the colonic mucosa,and the expression of Pxr mRNA,NF-κB p65 mRNA,and IκB-α mRNA.Results Compared with the blank control group,the INF-γ levels of in the UC rat model group,SASP group and each dose groups of Yuxian Decoction were significantly increased,and the level of IL-10 were significantly reduced(P<0.05);after treating with Yuxian Decoction,with the gradual increase of the administered dose,the INF-γ level was significantly reduced(P<0.05),and the IL-10 level was significantly increased(P<0.05),and the levels of IL-10 and INF-γ of the high-dose Yuxian Decoction group were significantly increased.There was no significant difference compared with the positive control SASP group(P>0.05).Compared with the blank control group,the expression level of Pxr and IκB-α mRNA were significantly reduced in the model group,and the expression of Pxr and IκB-α mRNA were signif
