摘 要:目的基于JAK2-STAT3信号通路研究祛脂愈肝汤对非酒精性脂肪性肝炎(NASH)大鼠白细胞介素(IL-17、IL-2)的影响。方法通过高脂饮食诱导复制NASH模型大鼠,随机均分为模型组、多烯组、祛脂愈肝汤低、中、高剂量组;另设空白组。经药物干预后、处死并观察肝组织病理变化;检测各组大鼠血清肝功能、血脂;ELISA法检测各组大鼠血清IL-17、IL-2水平;qRT-PCR法检测各组大鼠肝组织中JAK2、STAT3 mRNA表达。结果与空白组相比,模型组大鼠肝组织脂肪变性、炎性细胞浸润征象显著;各药物干预组均有不同程度的改善,其中祛脂愈肝汤高剂量组、多烯组改善最为显著;肝功能、血脂结果显示模型组大鼠血清谷草转氨酶(AST)、谷丙转氨酶(ALT)、甘油三酯(TG)、总胆固醇(TC)水平与空白组相比显著升高,差异有统计学意义(P<0.01),各药物干预组均有不同程度降低,差异有统计学意义(P<0.01);ELISA法检测模型组大鼠血清IL-17与空白组相比显著升高、IL-2显著降低,差异有统计学意义(P<0.01),各药物干预组血清IL-17显著降低、IL-2显著升高,差异有统计学意义(P<0.01);qRT-PCR法检测模型组大鼠肝组织JAK2、STAT3 mRNA表达与空白组相比显著升高,差异有统计学意义(P<0.01),各药物干预组肝组织JAK2、STAT3 mRNA表达显著降低,差异有统计学意义(P<0.01);综合上述结果,以多烯组、祛脂愈肝汤高剂量组疗效最佳,差异有统计学意义(P<0.05或P<0.01)。结论祛脂愈肝汤干预NASH模型大鼠,可通过抑制JAK2及下游STAT3 mRNA的表达,阻断JAK2-STAT3通路的激活,从而调控炎症介质的基因转录达到治疗NASH的作用。Objective The effects of Quzhi Yugan Decoction(祛脂愈肝汤)on interleukin-17(IL-17)and interleukin-2(IL-2)in non-alcoholic fatty liver disease(NASH)rats was studied based on Janus kinase 2(JAK2)and transcriptional activator protein 3(STAT3)signaling pathway.Methods NASH rats were induced by high-fat diet and randomly divided into the model group,the polyene phosphatidylcholine group,and the Quzhi Yugan Decoction low-dose,medium-dose and high-dose groups.And another blank group was set up.All the rats were sacrificed after drug intervention,stained with hematoxylin-eosin(H-E)to observe the pathological changes.Serum liver function and blood lipid of rats in each group were detected.Serum levels of IL-17 and IL-2 were determined by ELISA.Real Time-Quantitative Polymerase Chain Reaction(qRT-PCR)was used to detect the expressions of JAK2 and STAT3 mRNA in liver tissue.Results Compared with the blank group,the signs of steatosis and inflammatory cell infiltration in the liver tissue of rats in the model group were significant.All the drug intervention groups had different degrees of improvement,especially the high-dose group and polyene phosphatidyl choline group.The results showed that serum levels of aspartate aminotransferase(AST),alanine transaminase(ALT),triglyceride(TG)and total cholesterol(TC)in the model group were significantly increased compared with those of the blank group(P<0.01).After drug intervention,serum levels of AST,ALT,TG and TC in each intervention group were significantly reduced(P<0.01).Compared with those of the blank group,the level of serum IL-17 was increased significantly and serum IL-2 was significantly decreased in the model group by ELISA(P<0.01).After drug intervention,the serum IL-17 was significantly decreased and serum IL-2 was significantly increased in each intervention group(P<0.01).The expressions of JAK2 and STAT3 mRNA in liver tissues of rats in the model group were significantly increased compared with those of the blank group(P<0.01).After drug intervention,the expressio
