摘  要:目的 本研究拟在应用转录组测序得到的数据,探讨链球菌感染加重银屑病的可能基因。方法 基于链球菌感染与银屑病数据集(GSE158163, GSE14905),进行差异基因及功能通路富集分析。基于加权基因共表达网络分析(WGCNA)找到银屑病及链球菌感染的关键模块,找出基于WGCNA的两个数据集的差异基因,并通过蛋白功能相似性筛选出排名前五的基因。随后构建出mRNA-miRNA关系网络。结果 GSE158163数据集筛选出上调基因1 614个,下调基因1 614个,GSE14905数据集筛选出上调基因1 568个,下调基因1 699个,得出271个交集基因;功能富集到炎症反应调节、白细胞活化、含胶原蛋白的细胞外基质等通路;随后构建WGCNA网络,得出37个交集基因与链球菌感染与银屑病相关;将这37个基因与之前差异表达的271个基因共同筛选,筛选出9个交集基因;在这9个基因中根据其表达蛋白功能的差异进一步筛选出排名前五的基因,分别为S100A9、S100A12、S100A8、LRG1以及TGFA。结论 本研究基于公共数据库筛选出了5个链球菌感染和银屑病相关的基因,并论述了这些基因的病理生理机制,推测它们对两个疾病的影响。OBJECTIVE To explore the possible genes related to streptococcal infection and aggravated psoriasis based on the data obtained from transcriptome sequencing.METHODS Differential genes and enrichment of func-tional pathways were analyzed based on the dataset of streptococcal infection and psoriasis(GSE158163,GSE14905).The key modules of psoriasis and streptococcal infection were identified based on weighted gene co-expression network analysis(WGCNA),the differential genes were found out from the two datasets based on WGCNA,and the top 5 genes were screened out by means of similarity of protein function.The mRNA-miRNA network was constructed.RESULTS Totally 1614 up-regulated genes and 1614 down-regulated genes were screened out from GSE158163 dataset;totally 1568 up-regulated genes,1699 down-regulated genes and 271 in-tersected genes were screened from GSE14905 dataset.The functions were enriched in the pathways such as regu-lation of inflammatory response,leukocyte activation and collagen-containing extracellular matrix.The WGCNA network was constructed,and it concluded that 37 intersected genes were associated with the streptococcal infection and psoriasis.A common screening was carried out for the 37 genes and the 271 differentially expressed genes,and 9 intersected genes were screened out.The top 5 genes were further screened out from the 9 genes on basis of the difference of function of protein expression and were respectively as follows:S100A9,S100A12,S100A8,LRG1 and TGFA.CONCLUSION The 5 genes that are screened out from public database are associated with the streptococcal infection and psoriasis,the pathophysiological mechanisms of the genes are discussed,and the genes may have affections on the two diseases.
