摘  要:目的 应用生物信息学方法预测并分析刚地弓形虫顶端表面蛋白2(Toxoplasma gondii rhoptry apical surface protein 2,TgRASP2)的结构与功能,为刚地弓形虫的致病机制、药物靶点以及为疫苗的研发提供理论依据,方法 通过ToxoDB官网收集数据确定TgRASP2核酸序列和氨基酸序列;利用各类生物信息学在线网站分别对TgRASP2进行蛋白质基本理化性质、亲/疏水性、磷酸化位点、二级结构、三级结构以及抗原表位和抗原指数等进行预测和分析。结果 TgRASP2蛋白共含有437个氨基酸,分子式为C2185H3507N635O642S23,分子质量为49.682 13 ku,理论等电点为9.30,半衰期为30 h;蛋白不稳定系数为50.96,脂溶性指数为74.05,总平均亲水性系数为-0.620,属于亲水性蛋白;该蛋白无信号肽切割位点,无跨膜结构域,含有40个磷酸化位点;二级结构中α螺旋和延长链分别占34.10%和19.45%,β转角和无规则卷曲分别占7.55%和38.90%。经SMART网站分析,该蛋白的结构域含有4个低复杂区,无特殊结构域。该蛋白的抗原性为0.6651,共含有15个B细胞抗原表位,18个CTL抗原表位,15个Th抗原表位。结论 生物信息学预测TgRASP2蛋白具有亲水性,为可溶性蛋白。该蛋白存在多个潜在的B、T细胞抗原表位,具有免疫原性和反应原性,可作为弓形虫病疫苗候选蛋白。Objective In order to provide a theoretical basis for the pathogenicity and drug targets of Toxoplasma gondii and for the research and development of a vaccine,bioinformatics methods were applied to predict and analyze the structure and function of Toxoplasma gondii rhoptry apical surface protein 2(TgRASP2).Methods thedata were col-lected through the ToxoDB website to determine the nucleic acid sequence and amino acid sequence of TgRASP2;various bioinformatics online sites were used to predict and analyze the basic physicochemical properties of the protein,hydrophi-licity/hydrophobicity,phosphorylation sites,secondary structure,tertiary structure,as well as antigenic epitopes and anti-genic indices of TgRASP2,etc.,respectively.Results The results show that TgRASP2protein contains a total of 437a-mino acids,the molecular formula of C 2185 H 3507 N 635 O 642 S 23,the relative molecular mass of 49.68213ku,the theoretical i-soelectric point of 9.30,the half-life of 30h,the instability coefficient of 50.96,the lipid solubility index of 74.05and the o-verall average hydrophilicity coefficient of-0.620,a hydrophilic protein.The protein has no signal peptide cleavage site and no transmembrane structural domain,and contains 40phosphorylation sites(25serine phosphorylation sites,10threonine phosphorylation sites,5tyrosine phosphorylation sites);theα-helix and extended strand account for 34.10%and 19.45%of the secondary structure,while theβ-turned and random coiled account for 7.55%and 38.90%the secondary structure,respectively;After analysis on the SMART website,the structural domains of this protein contain four low-complexity re-gions and non-specific structural domains.The protein has an antigenicity of 0.6651and contains 15B-cell antigenic epitopes,18CTL cell antigenic epitopes and 15Th cell antigenic epitopes.Conclusion Bioinformatics predicts that the TgRASP2protein is hydrophilic and soluble.The presence of multiple potential B and T cell antigenic epitopes,immuno-genicity and reactogenicity of this protein can be
